To follow this, blood samples of 93 dogs were obtained from diverse areas of Iran and, in addition, 655 sequences of canine D-loop region obtained from GenBank. Total DNA of the samples were extracted by salting out procedure and applied as template for amplification and sequencing of the D-loop region of mtDNA. Sequence analysis of the 582-bp D-loop region of mtDNA in all samples defined a total of 25 haplotypes. The total nucleotide diversity was 0.01354± 0.00704 for Iranian native dogs. Analysis of molecular variance (AMOVA) showed significant percent for the variance between the studied populations (ΦST=0.029, P=0.0000). Fixation index values using F-Statistics method ranged from -0.02671 to 0.3748. Analysis of molecular variance displayed significant difference between some of these populations. The variation was especially observed between Tazi and Sarabi, Bakhtiari, Alborz native and Western regions populations or between Sarabi population with Kurdi, Sangsari, Alborz native and Western regions populations. Bakhtiari population showed a significant difference (P > 0.05) with Alborz native and Western regions populations, and finally European breeds exhibited a difference with South West Asia breeds. In addition Tazi, Sangsari and Western regions populations displayed significant difference with European breeds and South West Asia breeds (P > 0.05). Overall, the results of this study showed an acceptable genetic variation and genetic similarity in some Iranian dog populations.}, keywords = {Genetic structure- Mitochondrial- Iranian native dog,haplotype}, title_fa = {بررسی ساختار ژنتیکی سگ های بومی ایران با استفاده از اطلاعات ژنوم میتوکندری}, abstract_fa = {هدف تحقیق حاضر مطالعه ساختار ژنتیکی سگ­های بومی ایران و مقایسه با دیگر نژادهای خارجی است. برای این منظور نمونه­های خون از 93 قلاده سگ از جمعیت­های مختلف، از نقاط مختلف کشور جمع آوری گردید، همچنین به منظور مقایسه نتایج حاصل با دیگر نژادهای آسیایی و اروپایی، 655 توالی 582 جفت بازی از ناحیه کنترل ژنوم میتوکندری از بانک اطلاعات ژنوم دریافت شد. استخراج DNA کلیة نمونه­های خون صورت گرفت و برای تکثیر و تعیین توالی ناحیة کنترل ژنوم میتوکندری استفاده شد. با مطالعه توالی 582 جفت بازی ناحیة کنترل ژنوم میتوکندری، 25 هاپلوتیپ تشخیص داده شد. مقدار تنوع نوکلئوتیدی کل برای سگ­های بومی ایران 00704/0±01354/0 بدست آمد. نتایج آنالیز واریانس مولکولی با استفاده از روش F-Statistics مقدار معنی­داری را نشان داد (029/0=Fst و 000/0=P). مقادیر شاخص تثبیت با استفاده از این روش دارای دامنه­ای بین 02671/0- الی 37481/0 بودند. بر اساس این آزمون برخی از این جمعیت­ها بخصوص جمعیت تازی با جمعیت­های سرابی، بختیاری، بومی البرز و بومی غرب کشور، جمعیت سرابی با جمعیت­های کردی، سنگسری، البرز و بومی غرب کشور، جمعیت بختیاری با جمعیت­های بومی البرز و بومی غرب کشور و جمعیت اروپا با غرب آسیا با یکدیگر از نظر ژنتیکی متفاوت بودند (05/0>P). همچنین جمعیت­های تازی، سرابی و بومی غرب کشور با نژادهای اروپا و غرب آسیا اختلاف معنی­داری نشان دادند (05/0>P). جمعیت سگ­های بومی خراسان با همه جمعیت­های مورد بررسی شباهت ژنتیکی نشان داد (05/0}, keywords_fa = {سگ بومی ایران,تنوع ژنتیکی,ساختار ژنتیکی,هاپلوتیپ}, url = {https://jab.uk.ac.ir/article_1222.html}, eprint = {https://jab.uk.ac.ir/article_1222_579b614789613fe71c117e3fa2a0e704.pdf} }
