In order to identify the wild hosts of the virus, watermelon growing areas of Minab (Hormozgan province), Roodbar-e-Jonob and Arzuiyeh (Kerman province) were surveyed and weed samples collected within and around the severely infected farms. The infection of the samples was tested by PCR using specific primer pair. Results showed that 14 weed species from 12 genera and nine plant families are infected with WmCSV. Most of the infected weeds were symptomless and did not show any specific symptoms. All the infected weed species in this study are reported for the first time as the WmCSV host in the world. In order to compare WmCSV isolates from weed species, the amplified 655 bp segment of coat protein (CP) gene from 11 infected weeds were cloned and sequenced. Comparison of nucleotide and amino acid sequences of 11 WmCSV isolates showed close similarities with each other and with the GenBank isolates. The closest GenBank isolate was the previously reported Iranian isolate from Bandar-Abbas (Hormozgan province) with 98/9-99/2 and 96/8-100% homologies for nucleotide and amino acid sequences, respectively. According to the results of this study, weed species are secondary hosts and serve as the reservoirs of the WmCSV. Thus, these weeds could be potentially important in epidemiology of the virus.}, keywords = {Geminivirus,Begomovirus,Watermelon chlorotic stunt virus,Bemisia tabaci,Whitefly,Weeds}, title_fa = {شناسائی میزبان های وحشی ویروس کوتولگی سبزرد هندوانه در جنوب و جنوب شرق ایران}, abstract_fa = {ویروس کوتولگی سبزرد هندوانه (Watermelon chlorotic stunt virus, WmCSV) یکی از مخرب ترین ویروس های هندوانه در جنوب و جنوب شرقی ایران است. به منظور شناسائی علف هرزهای میزبان این ویروس، از مزارع بشدت آلوده هندوانه واقع در میناب (استان هرمزگان)، رودبار جنوب (جیرفت) و ارزوئیه (استان کرمان) بازدید گردید و نمونه برداری از داخل و حاشیه مزارع انجام شد. آلودگی و عدم آلودگی نمونه ها با آزمون PCR و با استفاده از آغازگرهای اختصاصی تعیین گردید. نتایج بدست آمده آلودگی تعداد زیادی از علف های هرز شامل چهارده گونه از دوازده جنس و نه تیره گیاهی را به WmCSV به اثبات رساند، بدون آنکه علائم خاصی روی بیشتر آنها دیده شود. تمامی این علف های هرز بعنوان میزبان WmCSV برای اولین بار معرفی می گردند. به منظور مقایسه جدایه های WmCSV در علف های هرز آلوده، قطعه 655 جفت بازی مربوط به ژن پروتئین پوششی 11 جدایه مختلف ویروس، همسانه سازی و تعیین ترادف گردید. مقایسه ترادف های نوکلئوتیدی و آمینواسیدی این جدایه ها با یکدیگر و با جدایه های موجود در بانک جهانی ژن نشان داد که جدایه های مورد مطالعه شباهت زیادی با یکدیگر و با سایر جدایه ها دارند. نزدیکترین جدایه WmCSV به جدایه های علف های هرز مورد مطالعه، جدایه بندرعباس (استان هرمزگان) بود که قبلا از این منطقه گزارش شده بود و از نظر ترادف های نوکلئوتیدی و آمینواسیدی بترتیب 2/99٪-9/98 و 100٪-8/96 با جدایه های فوق شباهت داشت. این نتایج نشان می دهد که این گیاهان بعنوان منابع نگاهدارنده WmCSV عمل کرده و بطور بالقوه می توانند در اپیدمیولوژی ویروس نقش اساسی داشته باشند.}, keywords_fa = {جمینی ویروس,بگوموویروس,ویروس کوتولگی سبزرد هندوانه,Bemisia tabaci,سفیدبالک,علف های هرز}, url = {https://jab.uk.ac.ir/article_1289.html}, eprint = {https://jab.uk.ac.ir/article_1289_afea31b1aadc142a2a9b97db99455ba3.pdf} }
