Moreover, Iran has the richest germplasm of pistachio due to the origin of pistachio and the existence of large numbers of commercial pistachio cultivars. Nevertheless, few chromosomal studies in pistachio have been yet reported and they however showed contradicting results on the number of chromosomes. In the present study, different pistachio species including P. vera, and wild species, i.e., Pistacia khinjuk and P. atlantica subsp. mutica were studied by classical chromosome staining and GISH technique. In order to stain chromosomes, two methods of aceto-iron-haematoxylin and Feulgen were used. Results indicated that the best staining was achieved by Feulgen compared with another method. Chromosome numbers of ‘Ohadi’, ‘Akbary’, ‘Ahmad-Aghaei’, ‘Nish-Kelaghi’ and ‘Khanjary’ were 2n=30 while chromosome numbers of ‘Italiaei’, ‘Ghazvini’ and ‘Sarakhs’ wild variety were 2n=28. Chromosome numbers of both P. khinjuk and P. atlantica subsp. mutica were 2n=28. In GISH experiment, no signal of hybridization of P. khinjuk and P. atlantica subsp mutica genomic probes were detected on P. vera chromosomes. This lack of detectable hybridization sites may however indicate that these chromosomes are probably originated from different taxa.}, keywords = {Pistachio,chromosome,Aceto-iron haematoxylin,Feulgen,GISH}, title_fa = {مطالعه سیتوژنتیک و بهینه سازی روش دورگ سازی ژنومی در محل در جنس پسته (Pistacia spp.)}, abstract_fa = {پسته (Pistacia vera) یکی از مهمترین محصولات باغی شناخته شده ایران در دنیا می‌باشد که نام ایران را با داشتن بیشترین ارقام تجاری دنیا به عنوان مهمترین مبنع ژرم پلاسم این گیاه مطرح نموده است. با این وجود تعداد مطالعات کروموزومی روی این محصول محدود و گزارشات موجود حاکی از تناقضاتی در تعداد کروموزوم‌ها می‌باشد. در این تحقیق گونه‌های پسته موجود در ایران شامل ارقام اهلی و گونه‌های غیر اهلی خنجوک و بنه با استفاده از روش‌های کلاسیک رنگ‌آمیزی کروموزوم و روش دورگ سازی ژنومی در محل مورد مطالعه قرار گرفتند. دو روش استو-آهن هماتوکسیلین و فولجن برای رنگ‌آمیزی کروموزوم‌ها به کار گرفته شد. بر اساس نتایج، رنگ‌آمیزی با فولجن، روش مناسب‌تری برای رنگ‌آمیزی کروموزوم‌های پسته در مقایسه با روش دیگر بود. تعداد کروموزم‌ها در ارقام اوحدی، اکبری، احمدآقایی، نیش کلاغی و خنجری دامغان 15 جفت (30=n2) در حالیکه ارقام ایتالیایی و قزوینی، واریته وحشی سرخس و گونه‌های خنجوک و بنه دارای 14 جفت کروموزوم (28=n2) بودند. در آزمایش دورگ سازی ژنومی در محل، تابش فلورسنتی که دال بر هیبریداسیون پروب‌های ژنومی خنجوک و بنه با نمونه‌های کروموزومی ارقام اهلی باشد، مشاهده نگردید. عدم مشاهده هیبریداسیون قابل ردیابی می‌تواند ناشی از مشتق شدن این کروموزوم‌ها از رده‌های مختلف باشد.}, keywords_fa = {پسته,کروموزوم,استو-آهن هماتوکسیلین,فولجن,دورگ سازی در محل}, url = {https://jab.uk.ac.ir/article_1333.html}, eprint = {https://jab.uk.ac.ir/article_1333_cc9dd9a834ed17380947493b42f5407b.pdf} }
