The PCR reactions were successfully performed with all primers. The results showed that all loci were quite polymorph. Hardy-Weinberg equilibrium test using chi-square test showed that some various combinations of loci-population were in a state of Hardy-Weinberg disequilibrium (P<0.05). The highest number of alleles was observed in loci of McMA2, McMA26 and OarHH35 of Dalagh, Arabic, Lory and Karakul populations respectively (12 alleles) and the lowest number of alleles for OarHH35 in Dalagh population. The maximum expected heterozygosity in combination locus-population of loci McMA2، McMA26، OarHH35، BM6444 were 0.885, 0.9, 0.88 and 0.87 respectively. With respect to the studied loci, it is concluded that all studied sheep populations have wide genetic diversity.}, keywords = {sheep,microsatellite markers,Genetic variation,Heterozygosity}, title_fa = {بررسی تنوع ژنتیکی پنج جمعیت گوسفند ایرانی با استفاده از نشانگرهای ریزماهواره‌ای}, abstract_fa = {نژادهای بومی در هر کشور به‌عنوان یک سرمایه ملی و محصولی استراتژیک در اقتصاد و رونق آن کشور محسوب می‌شوند که حفظ و نگهداری این نژادها بسیار ارزشمند است. با توجه به اهمیت حفظ تنوع در نژادهای بومی و شناسایی ذخایر ژنتیکی، پنج جمعیت از نژادهای گوسفند ایرانی (عربی، آرمان، دالاق، قره­گل و لری) با استفاده از 4 نشانگر ریزماهواره­ای (McMA2، McMA26، OarHH35 و BM6444) از نظر تنوع ژنتیکی مورد بررسی قرار گرفت. از تعداد 225 نمونه خون DNA ژنومی به روش استخراج نمکی بهینه‌شده، به دست آمد. واکنش‌های PCR به ‌خوبی انجام شد. نتایج نشان داد که تمامی جایگاه­ها کاملاً چندشکل بودند. آزمون تعادل هاردی–وینبرگ به روش آزمون مربع کای نشان داد که برخی ترکیبات مختلف جایگاه­ها-جمعیت در حالت عدم تعادل قرار داشتند (05/0P<). بیشترین تعداد آلل مشاهده ‌شده در جایگاه­های McMA2، McMA26 و OarHH35 به ترتیب مربوط به جمعیت­های دالاق، عربی، لری و قره­گل (12 آلل) و کمترین تعداد از آن جایگاه OarHH35 در جمعیت دالاق بود. حداکثر هتروزیگوسیتی مورد انتظار در حالت ترکیب جمعیت-جایگاه برای جایگاه­های McMA2، McMA26، OarHH35، BM6444 به ترتیب 885/0، 9/0، 88/0و 87/0 بودند. در مجموع می­توان نتیجه گرفت که جمعیت­های گوسفند مورد بررسی در این پژوهش با توجه به جایگاه­های مورد مطالعه، از تنوع ژنتیکی نسبتاً بالایی برخوردارند.}, keywords_fa = {نشانگرهای ریزماهواره‌ای,تنوع ژنتیکی,چندشکلی}, url = {https://jab.uk.ac.ir/article_1396.html}, eprint = {https://jab.uk.ac.ir/article_1396_8ebbb6214a7d04c1d4ad7dec40b90bd6.pdf} }
