In present study, activity of defensive enzymes such as alternative oxidase (AOX), superoxide dismutase (SOD), catalase (CAT), electrolyte leakage index (ELI) and hydrogen peroxide (H2O2) amount as a cellular damage indices in two genotypes of chickpea, susceptible (ILC533) and tolerant (Sel96Th11439) under 4°C cold stress was  evaluated. Statistical analyses results showed that there is a significant difference between genotypes under cold stress. Under cold stress, decrease in damage indices was accompanied with an increase in AOX activity. The maximum activity of AOX was seen in tolerant genotype on sixth day of cold stress, whereas under such conditions AOX activity in susceptible genotype significantly was lower compared to tolerant genotype. The increase in AOX activity which was accompanied with a decrease in cellular damage (ELI and H2O2 results) particularly on sixth day of cold stress showed the significance of AOX enzyme in tolerance of chickpea plants to cold stress. The simultaneous and significant increase in SOD and CAT activities along with confirmation the results of AOX activity increased degree of genetic tolerance of chickpea to cold stress or plant recovery after exposure to cold stress. These indices may be useful in assessment of chickpea genotypes under cold stress or utilization them in breeding programs.}, keywords = {alternative oxidase,cold,electrolyte leakage,Chickpea}, title_fa = {ارزیابی فعالیت برخی از آنزیم‌های دفاعی نخود (Cicer arietinum L.) تحت تنش سرما}, abstract_fa = {گیاهان جهت بقا و تحمل تنش‌های محیطی نیازمند مکانیسم‌های دفاعی متعددی هستند. در این پژوهش فعالیت آنزیم‌های اکسیداز متناوب (AOX) میتوکندری، سوپراکسید دیسموتاز (SOD) و کاتالاز (CAT) ، میزان نشت الکترولیتی (ELI) و پراکسید هیدروژن (H2O2) به عنوان شاخص‌های خسارت سلولی در دو ژنوتیپ حساس (ILC533)  و متحمل (Sel 96TH11439) نخود زراعی (Cicer arietinum L.) تحت تنش سرمای 4 درجه سانتی‌گراد ارزیابی شد. نتایج تجزیه آماری نشان داد که اختلاف معنی‌داری بین ژنوتیپ‌ها تحت تیمارهای دمایی وجود داشت. تحت تنش سرما به موازات کاهش شاخص‌های خسارت، فعالیت آنزیم AOX افزایش معنی‌داری یافت. بیشترین فعالیت این آنزیم در ژنوتیپ متحمل در روز ششم پس از تنش سرما مشاهده شد در حالی­که تحت این شرایط میزان فعالیت این آنزیم در ژنوتیپ حساس به­طور معنی­داری کمتر از ژنوتیپ متحمل بود. افزایش فعالیت آنزیم AOX که با خسارت سلولی کمتر (نتایج ELI و H2O2) به­خصوص در روز ششم پس از تنش سرما همراه بود، بیانگر اهمیت این آنزیم در تحمل به سرما در گیاه نخود می‌باشد. افزایش معنی­دار و همزمان الگوی فعالیت آنزیم‌های SOD و CAT ضمن تایید نتایج فعالیت AOX، درجه تحمل ژنتیکی نخود به سرما را افزایش داده و یا اینکه سبب بهبود گیاه پس از اعمال تنش می‌شود. چنین شاخص‌هایی ممکن است در ارزیابی ژنوتیپ‌های نخود تحت تنش سرما و یا بکارگیری آن‌ها در برنامه‌های اصلاحی مفید باشند.}, keywords_fa = {اکسیداز متناوب,سرما,نشت الکترولیتی,نخود}, url = {https://jab.uk.ac.ir/article_1635.html}, eprint = {https://jab.uk.ac.ir/article_1635_7cc06d8f83029aa8ed91476a9fdaf55f.pdf} }
