This glucosinolate is found in huge scale in Lepidium draba plant from the brassicaceae family.   Materials and Methods: In this research, sulforaphane production and the gene expression level of CYP79F1 (one of the critical enzyme in glucoraphanin biosynthesis pathway) and myrosinase were investigated in Lepidium draba seedlings under treatment with different concentrations of ZnO (0, as a control, 25, 100, 500 and 1000 mg/L).   Results  The results showed that sulforaphane content in treated seedling significantly increased in comparison with the control in the presence doses more than 100 mg/L ZnO. Based on the results, gene expression level of CYP79F1 decreased by the increase of ZnO concentration in media, and the increment was significant at the 5% level in the presence of the highest dose in compared to the control. In contrast, the myrosinase   gene expression level evaluated by increasing ZnO concentrations in media and the increment was significantly at the doses more than 500 mg/L ZnO concentration.   Conclusions According to the results, it seems that evaluating of the sulforaphane production level in the treated seedlings is more attributed to the increased gene expression level of myrosinase which resulted in the enhanced enzyme production.}, keywords = {Gene expression,Myrosinase,Soulforaphane,ZnO}, title_fa = {محتوای سولفورافان و بیان ژن‌های CYP79F1 و میروزیناز در گیاهچه‌های تیمار شده ازمک با غلظت‌های مختلف اکسید روی}, abstract_fa = {هدف: سولفورافان یک ایزوتیوسیانات مشتق شده از گلوکوزینولات گلوکورافانین تحت  هیدرولیز آنزیمی میروزیناز می­باشد که اثرات درمانی متعددی دارد. این گلوکوزینولات به فراونی در گیاه ازمک از خانواده براسیکاسه یافت می­شود. مواد و روش­ها: در این تحقیق، تولید سولفورافان، بیان ژن‌ CYP79F1 (یکی از آنزیم­های کلیدی در بیوسنتز گلوکورافانین) و بیان ژن میروزیناز در گیاهچه­های ازمکتیمار شده با غلظت‌های مختلف (صفر (به عنوان شاهد)، 25، ، 100، ، 500 و 1000 mg/L ) اکسید روی بررسی شد. نتایج: نتایج نشان داد که محتوای سولفورافان گیاهچه­های تیمار شده بصورت معنی­داری نسبت به شاهد افزایش (در غلظت­های بالاتر از 100 mg/L) یافته است. بر اساس نتایج حاصله، بیان ژن CYP79F1 با افزایش غلظت اکسید روی در محیط به صورت خطی کاهش نشان داد و این کاهش در حضور بالاترین غلظت، در سطح 5 درصد نسبت به نمونه شاهد معنی­دار می­باشد. در مقابل، بیان ژن آنزیم میروزیناز با افزایش غلظت اکسید روی در محیط به صورت گرادیان افزایش نشان داد و این افزایش در حضور غلظت­های بالاتر از mg/L 500 نسبت به نمونه شاهد معنی­دار بود. نتیجه­گیری: بر اساس نتایج، چنین بنظر می­رسد که افزایش تولید سولفورافان در گیاهچه­های تحت تیمار، بیشتر به دلیل افزایش بیان و در نتیجه تولید بیشتر آنزیم میروزیناز بوده است.}, keywords_fa = {اکسید روی,بیان ژن,میروزیناز,سولفورافان}, url = {https://jab.uk.ac.ir/article_2474.html}, eprint = {https://jab.uk.ac.ir/article_2474_bd569483a774b16bdbaf2217b889ea5a.pdf} }
