Ethylene is one of the major factors that limits post-harvest longevity. Therefore, genetic manipulation has been considered to reduce the adverse effects of this hormone and the present research investigates transgenic roses containing the mutant etr1-1 gene. In addition, according to antagonist effects, the change of gibberellin concentration to ethylene in favor of higher gibberellin concentration in the present research has been studied.       Materials and methods The content of phytohormones in long-lasting line (transgenic) and control (non-transgenic) after application of ethylene (0 and 1 µL L-1) and gibberellin (80 mg L-1) treatments were investigated in bud and half-open stages. Application of ethylene treatment was performed by injecting pure gas into polyethylene bags by syringe. For the application of gibberellin treatment, cut flowers were placed in gibberellin solution at the desired concentration. The treatments were applied for 24 hours and samples were taken from the outermost ring of petals. Experiments were conducted as factorial in a completely randomized design with four replications in order to measure phytohormones.   Results The results showed a decrease in the amount of gibberellin, benzyl adenine and indole acetic acid hormones and an increase in the amount of abscisic acid and ethylene hormones in bud and half-open stages in long-lasting line were significant compared to the control. In addition, the highest content of gibberellin, indole acetic acid and benzyl adenine hormones and the lowest content of ethylene and abscisic acid belongs to the GA3-treated long-lasting line.   Conclusion Therefore, etr1-1 gene appears to be the right candidate for delaying ethylene-dependent aging in sensitive flowers and with GA3 treatment could be considerably improved longevity by reduction of oxidative damage. }, keywords = {Cut rose,Ethylene,Genetic manipulation,Gibberellin}, title_fa = {بررسی میزان هورمون‌های گل سرخ ترا‌ریخته با ژن etr1-1 تحت تیمارهای اتیلن و جیبرلین}, abstract_fa = {هدف: گل سرخ از مهم­ترین گل­های شاخه بریده دنیا بوده که تا کنون مطالعات زیادی برای حفظ کیفیت و ماندگاری پس از برداشت آن انجام شده است. از عوامل اصلی محدود کننده ماندگاری در مرحله پس از برداشت می­توان به هورمون اتیلن اشاره نمود. لذا دست­ورزی ژنتیکی به ­منظور کاهش اثرات نامطلوب این هورمون مورد توجه بوده و در پژوهش حاضر، گل­های سرخ تراریخته حاوی ژن جهش­یافته etr1-1 مورد ارزیابی قرار گرفتند. همچنین با توجه به اثرات آنتاگونیستی، تغییر غلظت هورمون جیبرلین نسبت به اتیلن به نفع بالاتر بردن غلظت جیبرلین در مطالعه اخیر مورد توجه قرار گرفت. مواد و روش­ها: میزان فیتوهورمون­های لاین­ ماندگار (تراریخته) و شاهد (غیر تراریخته) پس از اعمال تیمارهای اتیلن (صفر وL L-1µ 1) و جیبرلین (mg L-1 ۸۰) در مراحل غنچه تجاری و نیمه باز بررسی شدند. اعمال تیمار اتیلن با تزریق گاز خالص به داخل کیسه­های پلی­اتیلنی توسط سرنگ انجام شد. جهت اعمال تیمار جیبرلین، گل­های شاخه بریده در محلول جیبرلین با غلظت مورد نظر قرار داده شدند. تیمارها به مدت ۲۴ ساعت اعمال گردید و نمونه­برداری از بیرونی­ترین ردیف گلبرگ­ها انجام شد. آزمایش­ها به­صورت فاکتوریل در قالب طرح کاملاً تصادفی با چهار تکرار به منظور اندازه­گیری میزان فیتوهورمون­ها اجرا شد. نتایج: نتایج نشان داد که کاهش در میزان هورمون­های جیبرلین، بنزیل آدنین و ایندول استیک اسید و افزایش در میزان هورمون آبسیزیک اسید و اتیلن در مراحل غنچه و نیمه­باز در لاین ماندگار نسبت به شاهد معنی­دار بود. همچنین، بیشترین میزان هورمون­های جیبرلین، بنزیل آدنین و ایندول استیک اسید و کمترین میزان اتیلن و آبسیزیک اسید مربوط به لاین ماندگار در تیمار با جیبرلین بود. نتیجه­گیری: با توجه به نتایج موجود به نظر می­رسد که ژن etr1-1 می­تواند کاندیدای مناسبی، جهت به تأخیر انداختن فرایند پیری وابسته به اتیلن در گل­های حساس باشد که همراه با تیمار جیبرلین از طریق کاهش خسارت اکسیداتیو ماندگاری را به­طور قابل توجهی افزایش می­دهد.}, keywords_fa = {اتیلن,جیبرلین,دست‌ورزی ژنتیکی,گل سرخ شاخه بریده}, url = {https://jab.uk.ac.ir/article_2670.html}, eprint = {https://jab.uk.ac.ir/article_2670_9b5db01ca174177ea49cfbfbf2235d81.pdf} }
