Thermal stressors such as heat stress elicit neuroendocrine responses including activation of the HPA axis .Heat stress is a challenge for animal and human society. It has been shown that brain monoaminergic system is changed during acute and chronic heat stress condition. Commercial and domestic poultry are very susceptible animals to the heat stress. High environmental temperature led to heat stress condition and had negative effect on physiological performance. One effect of heat stress is changed molecular and cellular mechanisms. These changes in diencephalon, as an awaking center (thalamus) and autonomic mechanisms center (hypothalamus), have profound physiological performance results. However the goal of experiment was to investigate effect of heat stress on diencephalic BDNF gene expression and blood corticostrone level.     Materials and Methods In this experiment, 20 seven-day-old male chicks divided in 1- heat stress group (10 chicks) and 2- control group (10 chicks in normal temperature condition). Heat stress induced by 39 C◦ and continued to 5 hours. Control group temperature was 30 C◦. At the end of 5 hours duration, chicks was anesthetized with Isofluran gas and killed. Blood gathered from jugular vein. Brain dissected and diencephalon was frozen in liquid nitrogen immediately. Blood corticosterone level was measured by ELIZA method, BDNF gene expression assayed by quantitative real time PCR technique with the SYBR green method.   Results In this experiment, heat stress significantly reduced diencephalic BDNF gene expression. However, corticosterone level significantly elevated in heat stress exposed chicks. It would be able to say that there is a relation between BDNF gene expression and blood corticosterone concentration.   Conclusions These results showed changes in BDNF gene expression during heat stress condition is associated to blood corticosterone level elevation. Taken together, the results showed that a significant relationship between Hypothalamus-Hypophysis- Adernal cortex activity during acute heat stress condition and diencephalic BDNF gene expression as a neuroprotective agent of brain. }, keywords = {Broiler chick,Brain derived neurotrophic factor (BDNF),Corticosterone,Heat stress}, title_fa = {تاثیر استرس گرمایی بر سطح کورتیکواسترون خون و بیان ژن فاکتور رشد عصبی مشتق از مغز ناحیه دیانسفال جوجه 7 روزه نژاد گوشتی}, abstract_fa = {هدف: استرس گرمایی یکی از چالش­های جدی در جوامع بشری و حیوانات  می­باشد. استرس­ها با تغییر مکانیسم­های بدن سبب و زمینه ساز ایجاد بیماری­ها می­باشند. درجه حرارت بالای محیط، سبب ایجاد استرس حرارتی می­شود که اثرات منفی بر عملکرد فیزیولوژیک دارد. یکی از آثار نهان استرس، تغییر در مکانیسم­های سلولی و ملکولی می­باشد. این تغییرات در دیانسفال به دلیل اهمیت این ناحیه در کنترل اطلاعات حسی ورودی به مغز (تالاموس) و مکانیسم­های خودکار بدن (هیپوتالاموس) نتایج عمیقی در عملکرد حیوان دارد. از این رو هدف آزمایش بررسی تاثیر استرس گرمایی حاد بر بیان ژن BDNF[1] و سطح کورتیکواسترون خون بود. مواد و روش‌ها: در این آزمایش تعداد 20 جوجه 7 روزه سن نر نژاد راس 308 به دو دسته تحت استرس گرمایی 39 درجه و دمای نرمال 30 درجه تقسیم و به مدت 5 ساعت تحت استرس گرمایی قرار گرفتند. در پایان جوجهها با گاز ایزوفلوران بیهوش، خونگیری از سیاهرگ وداج انجام و کشتار شده  و ناحیه دیانسفال آنها برداشته شد و برای آنالیز بیشتر در یخچال منفی 80 درجه سانتیگراد نگهداری شد.  میزان بیان ژن BDNF به روش ریل تایم کمّی و سایبر گرین اندازه گیری شد. سطح کورتیکواسترون خون با روش الایزا مورد سنجش قرار گرفت. نتایج: در این آزمایش استرس گرمایی به طور معنی­داری بیان ژن BDNF ناحیه دیانسفال را کاهش داد. اما سطح کورتیکواسترون خون جوجه های تحت استرس گرمایی به طور معنی­داری افزایش نشان داد. نتیجه‌گیری: این نتایج نشان می­دهد که بین سطح کورتیکواسترون خون و بیان ژن BDNF رابطه معکوس وجود دارد. با توجه به این می­توان رابطه مهمی میان استرس گرمایی و فعالیت محور هیپوتالاموس-هیپوفیز-قشر آدرنال مشاهده کرد که با تاثیر بر بیان ژن فاکتور عصبی مشتق از مغز سبب اختلال در نقش حفاظت نورونی این پروتئین در مغز می­گردد.
