Suitable climatic conditions for apricot production and its economic importance were the main reasons for its vast area under cultivation and diversity. Genetic diversity of 42 apricot cultivars and genotypes collected from new Iranian and Czech Republic collections was studied using fluorescent-AFLP markers. The objectives of this study were to determine the genetic structure and genotypic diversity among different eco-geographical populations. The results showed that five pairs of fluorescent-AFLP primers showed clear banding patterns and high level of polymorphism amongst the 30 pairs of primers screened. The total bands were 187 and MGAG ETG primer showed 63% polymorphism among primers that was the highest quantity of polymorphism. Based on the application of genotypes from breeding programs, Hybridization and selection, there were not appropriate results from UPGMA method. We used RB method to analyze the relationship among samples. Six groups of cultivars and genotypes were detected based on their origin and geographical distribution. Our findings showed that in comparison with traditional AFLP, fluorescent-AFLP markers are more efficient in evaluating genetic diversity and population structure among studied apricot genotypes and could be useful in other Prunus fruit tree species}, keywords = {Apricot,fluorescent AFLP-markers,polymorphism,genetic diversity}, title_fa = {مطالعه تنوع ژنتیکی و گروه های جغرافیایی برخی از ارقام و ژنوتیپ های زردآلو با استفاده از نشانگرAFLP}, abstract_fa = {زردآلو با نام علمی (Prunus armeniaca L.) متعلق به خانواده رزاسه است که بومی مناطق مدیترانه و آسیای میانه به ویژه ایران می باشد. هدف از این مطالعه ارزیابی تنوع ژنتیکی و ساختار ژنتیکی ارقام و ژنوتیپ های زردآلو بر اساس منشا جغرافیایی آنها می باشد. بر این اساس تنوع ژنتیکی 42 رقم و ژنوتیپ زردآلوی موجود در کلکسیون های سهند تبریز و باغ گیاهشناسی دانشکده باغبانی لدنیسه جمهوری چک، با استفاده از نشانگرهای AFLP نشاندار با فلورسنت ارزیابی شد. در ابتدا 30 ترکیب پرایمر انتخابی بر اساس ترکیب پرایمری موجود روی 42 نمونه زردآلو مورد آزمایش قرار گرفت که 5 ترکیب پرایمری با بیشترین مقدار چندشکلی و الگوی باندی واضح، در ادامه پژوهش مورد استفاده قرار گرفتند. تعداد کل باندهای محاسبه شده توسط این 5 پرایمر 187 بود که پرایمر MGAG/ETG با مقدار چندشکلی برابر با 1/63% بیشترین میزان چندشکلی و پرایمر MTTT/ETG با چندشکلی برابر با 9/53 % کمترین مقدار را نشان دادند. نتایج این پژوهش نشان داد ارقام و ژنوتیپ های زردآلو موجود در کلکسیون های ایران و چک بر اساس منشا و پراکندگی جغرافیایی در گروهبندی های مجزایی قرار می گیرند. بررسی تنوع ژنتیکی و ساختار جمعیت بین ارقام و ژنوتیپ های زردآلوی مورد مطالعه و حتی دیگر گونه های Prunus با استفاده از ترکیب نشانگرهای AFLP نشاندار در مقایسه با روش متداول AFLP نتایج کارآمدتری را نشان داد}, keywords_fa = {زردآلو,نشانگرهای AFLP نشاندار,چند شکلی,تنوع ژنتیکی}, url = {https://jab.uk.ac.ir/article_461.html}, eprint = {https://jab.uk.ac.ir/article_461_8549611008f2d38355434caca44a89fc.pdf} }
