همسانهسازی، بیش بیان و بررسی خصوصیات آنزیم قلیایی فیتاز (phyC) در باکتری اشرشیا کلی
فیتاز (مایواینوزیتول هگزا کیس فسفات فسفو هیدرولاز) آنزیمی هیدرولیزی است که فسفات معدنی تولید میکند. ژن کد کننده فیتاز phyC از باکتری باسیلوس سابتیلیس ATCC12711 جداسازی و توالی یابی شد. توالی نوکلوتیدی ژن فیتاز حاوی ناحیه کدکننده به طول 1089 جفت باز است که 90 نوکلوتید ابتدایی آن، مربوط به سیگنال پپتید ژن میباشد. به منظور تولید آنزیم فیتاز نوترکیب، ژن هدف به وکتور بیانی pET32a (+) وارد شد و وکتور نوترکیب پس از تکثیر در باکتری اشرشیا کلی DH5α به باکتری اشرشیا کلی BL21(DE3) به عنوان میزبان بیان منتقل شد. تحریک بیان ژن با استفاده از IPTG در غلظت نهایی 1 میلی مولار در دمای 30 درجه سانتیگراد و حضور کلرید کلسیم 10 میلیمولار صورت گرفت. نمونهگیری در زمانهای صفر تا پنج ساعت انجام شد و میزان تولید پروتئین نوترکیب با استفاده از الکتروفورز بررسی گردید. فیتاز محلول حاصل از ژن phyC-Trx با موفقیت به شکل درونسلولی در باکتری اشرشیا کلی بیان شد. وزن مولکولی فیتاز نوترکیب تولید شده در حدود 64 کیلو دالتون تخمین زده شد. اندازهگیری فعالیت آنزیمی با استفاده از روش استاندارد فسفاتاز، فعالیت آنزیم نوترکیب تولید شده را 44/7 واحد در میلیلیتر نشان داد. همچنین pH بهینه برای فعالیت آنزیم فیتاز قلیایی نوترکیب در حدود 7 برآورد شد. نتایج نشان داد که فیتاز بدست آمده از باکتری باسیلوس سابتیلیس به جهت پایداری حرارتی و همچنین هزینه پایین تولیدی گزینه مطلوبی جهت استفاده در صنعت میباشد.

استفاده از روش رگرسیون متغیر های ظاهری برای پیش بینی مقدار تولید شیر و چربی در جمعیت گاوهای هلشتاین ایران
ژن DGAT1 به عنوان یک ژن کاندید در کروموزوم شماره 14 برای درصد چربی و تولید شیر مطرح می باشد. این ژن با کد کردن آنزیم دی آسیل گلیسرول اسیل ترانسفراز نقش اصلی را در سنتز تری گلیسیرید و در نهایت چربی شیر دارد. در این پژوهش 398 نمونه خون از گاوهای هلشتاین استانهای تهران و اصفهان جمع آوری گردید، با انجام واکنش PCR یک قطعه 411 جفت بازی از اگزون شماره 8 این ژن تکثیر گردید. تعیین ژنوتیپ افراد جمعیت با استفاده از تکنیک PCR-RFLP انجام شد. بر اساس نحوه برش آنزیم CfrI سه نوع ژنوتیپ KK ،KA ،AA شناسایی شدند، همچنین فراوانی آللی K و A به ترتیب برابر با 37/0 و 63/0 تعیین شدند. سپس با استفاده از برازش GLM ، صفاتی که اثر ژن DGAT1 برا ی آنها معنی دار بود تعیین شدند و در نهایت با استفاده از کد بندی عامل های مدل و رگرسیون متغیر های ظاهری، مدل های پیش بینی ارائه شدند. از ضریب همبستگی بین رکورد های واقعی و پیش بینی شده به عنوان معیار اعتبار سنجی مدل های ارائه شده استفاده شد. نتایج این پژوهش نشان داد که مدل های ارائه شده برای پیش بینی صفات تولید شیر 305 روز، ارزش اصلاحی درصد چربی و درصد چربی بر اساس دو بار دوشش در روز، تا حدودی قادر به پیش بینی تولید می باشند اما به دلیل استفاده از حجم اطلاعات کم، مقدار عددی ضرایب همبستگی به میزان کمی برآورد شدند. با استفاده از اطلاعات ژن ها و رکورد های یبشتر می توان نتایج دقیق تری بدست آورد.

بررسی تنوع ژنتیکی ارقام توتون دسته بارلی و ویرجینیا با استفاده از چند شکلی طولی قطعات تکثیری
با توجه به اهمیت توتون از نظر اقتصادی و نقش آن به عنوان یک گیاه مدل در پژوهشهای مولکولی در پژوهش حاضر به بررسی میزان شباهت ژنتیکی 50 ژنوتیپ توتون از دو دسته بارلی و ویرجینیا پرداخته شد. به منظور تعیین ژنوتیپ از 21 ترکیب آغازگری نشانگر AFLP استفاده شد. از تعداد کل 480 نوار ایجاد شده، با تعداد متوسط 76/17 نوار در هر ترکیب، 373 نوار چندشکل بودند. با توجه به میانگین درصد چندشکلی بالا (45/77 درصد) میتوان انتظار داشت بتوان برای شناسایی تنوع ژنتیکی موجود در ارقام از این نشانگر به عنوان ابزاری قدرتمند در برنامههای اصلاحی توتون استفاده نمود. بررسی ضرایب ژنتیکی جاکارد نشان داد ژنوتیپهای Badisher Burley E، Pennbel69، R9 و Coker 176 واجد بالاترین فاصله ژنتیکی از بقیه ژنوتیپها هستند. لذا، به نظر میرسد بتوانند در برنامههای اصلاحی از جمله هیبریداسیون و تشکیل جمعیتهای در حال تفرق برای مکانیابی QTL ارزشمند باشند. نتایج تجزیه واریانس مولکولی نیز 4 درصد از کل تنوع ژنتیکی موجود در بین ژنوتیپها را تنوع بین دو دسته بارلی و ویرجینیای گرمخانهای برآورد نمود و 96 درصد از تنوع موجود را در درون دستهها نشان داد. همچنین بررسی آمارههای تنوع ژنتیکی نشان داد ترکیبات آغازگری E060-M160، E070-M140، E070-M150 و E070-M160، E080-M160، E080-M150، E100-M140 و E100-M150 درکل به عنوان قدرتمندترین نشانگرها از بین کل ترکیبات آغازگری در ارزیابی و تشخیص روابط بین ژنوتیپهای توتون مورد مطالعه هستند و میتوانند در سایر پژوهشهای توتون مورد توجه قرار گیرند.

تنوع آللی نشانگرهای ناحیهQTL کنترل کننده تحمل به شوری ( Saltol ) در مرحله گیاهچه ای در ارقام برنج ایرانی
با توجه به ضرورت تحمل به شوری در برنج و از طرفی وجود تحقیقات مختلف اصلاح نباتات مولکولی در شناسایی QTLهای بزرگ اثر کنترل کننده تحمل به شوری، ضرورت شناسایی حضور این QTLها در ارقام مختلف ایرانی و شناسایی تاثیرگذارترین QTLها در پیشبرد انتخاب در برنامههای اصلاحی بسیار مفید است. در این مطالعه، پاسخ به شوری 20 رقم مختلف برنج ایرانی و دو شاهد متحمل (Pokkali) و حساس (IR29 ) در مرحله گیاهچهای در شرایط گلخانه ( dsm-110 و 6 ، 0EC=) با استفاده از 6 نشانگر چند شکل ریزماهواره مورد ارزیابی قرار گرفت. نتایج تجزیه واریانس اختلاف بسیار معنیداری بین ژنوتیپها از نظر صفات مختلف نشان داد. ژنوتیپهای برنج از نظر تحمل به شوری پاسخ متفاوتی در مرحله گیاهچهای نشان دادند. دندروگرام به دست آمده از روش UPGMA (روش اتصال میانگین)، ارقام را به سه گروه طبقهبندی کرد. بیست رقم برنج بر اساس QTL بزرگاثر ناحیه Saltol برای تحمل به شوری بر روی کروموزوم 1 در گروه های هاپلوتیپی جداگانه قرار گرفتند. نتایج نشان دهنده پتانسیل ژرمپلاسم برنجهای ایرانی جهت یافتن ژنهای جدید تحمل در القاء تحمل به شوری، به غیر از ناحیه Saltol در مرحله گیاهچهای بود. همچنین ، نشانگر RM8094در مرحله گیاهچه ای به عنوان مؤثرترین نشانگر جهت شناسایی ژنوتیپ های متحمل به شوری میباشد.

ترانسفورماسیون گیاه شابیزک Atropa belladonna توسط Agrobacterium rhizogense
شابیزک (Atropa belladonna) یکی از مهمترین گیاهان دارویی خانواده سولاناسه می باشد. این گیاه حاوی منابع آلکالوئیدهای تروپانی است. این آلکالوئیدها بیشتر درریشه ها بیوسنتز می شوند. تحقیق حاضر به منظور افزایش ریشه های مویی شابیزک به روش انتقال ژن انجام شد. برای این منظور از سویه آگروباکتریوم رایزوژنز (15843 AR) استفاه شد. ابتدا ریزنمونه های ساقه با سوسپانسیون آگروباکتریوم دارای ژن rolB تلقیح شدند. سپس جهت باززایی مستقیم از پنج ترکیب (MS، KIN mg/l 3MS + ،B5، KIN mg/l 12 MS+ و mg/l KIN5 B5 +) در قالب طرح کاملاٌ تصادفی در سه تکرار استفاده شد. نتایج تجزیه آماری نشان داد که سطح هورمونی KIN mg/l 12+ MS بیشترین باززایی مستقیم از نوساقه را دارا میباشد. برای آنالیز گیاهان تراریخت از دو روش فنوتیپی (مشاهده ریشه مویی از گیاهچه تراریخت) و تکثیر بخشی از ژن rolB (به روش PCR) استفاده شد.

بررسی کال زایی و جنین زایی سوماتیکی در زیره سیاه (.Bunium persicum Boiss)
زیره سیاه (Bunium persicum Boiss.) گیاهی از خانواده چتریان است که علیرغم اهمیت دارویی فراوان، به خاطر مشکلات ناشی از جوانهزنی و طول دوره رویش طولانی، تاکنون در کشورمان اهلی نشده است و در طبیعت بهصورت وحشی میروید. تکنیکهایی نظیر کشت بافت، میتواند جهت کاهش طول دوره رویش در این گیاه بسیار مؤثر باشد. در این تحقیق بهینه سازی رشد جنین های سوماتیک، در جهت تولید و تجاری شدن این گیاه دارویی و تولید بذر مصنوعی مورد مطالعه قرار گرفت. از ریزنمونههای هیپوکوتیل و برگ کوتیلدونی اکوتیپ منطقه اصفهان و محیط کشت MS با 14 تیمار هورمونی مختلف جهت بررسی میزان تولید کالوس و جنین های سوماتیکی استفاده گردید. آزمایش بصورت فاکتوریل و بر پایه طرح کاملاً تصادفی چند مشاهدهای و در 4 تکرار اجرا گردید. بهترین ریزنمونه جهت تولید جنین سوماتیکی، هیپوکوتیل و بهترین تیمار هورمونی جهت کالوس زایی، ترکیب هورمونی mg/l Kin 5/0mg/l NAA+ 1 و mg/l NAA1 و جهت باززایی جنین رویشی mg/l 2ip2/0 + mg/l IBA01/0 +2/1MS می باشد. جنین های تولیدی در محیط کشت MS حاوی سیتوکینین 2ip و اکسین IBA رشد طبیعی نشان دادند. با توجه به مشکلات ناشی از جوانه زنی بذور در زیره سیاه، تولید بذر مصنوعی در آن از اهمیت ویژه ای برخوردار است. با توجه به موفقیت تولید جنین های سوماتیکی در این پژوهش، تولید بذر مصنوعی در زیره سیاه می تواند مورد بررسی قرار گیرد

بررسی پروتئوم پیام رسانی خشکی در ریشه برنج
خشکی یکی از بزرگترین عوامل محدودکننده عملکرد گیاهان در سطح جهان میباشد. بررسی تغییرات الگوی پروتیینهای ریشه برنج در سیستم خشکی نسبی با استفاده از تکنیک پروتئومیکس مورد مطالعه قرارگرفت. تیمارها در سه تکرار مستقل از هم اعمال شد و چهار بافت ریشهای مورد مقایسه قرارگرفت: ریشههایی که تحت آبیاری کامل قرارداشتند WW))، ریشه هایی که تحت خشکی کامل بوده (WD)، ریشه هایی که در تقسیم ریشه آب کافی دریافت کرده (SpWW) و ریشه هایی که در تقسیم ریشه در معرض خشکی کامل بودند (spWD) بررسی پروتئوم بافتهای ریشه حاصل از تیمارهای ذکر شده توسط الکتروفورز دوبعدی منجر به شناسایی 32 نقطه پروتیین شد که از این تعداد 13 نقطه پروتیینی در هر دو تیمار WD و spWD افزایش بیان داشته، تعداد 9 نقطه پروتیینی در هر سه تیمارWD، spWD و spWW نسبت به گیاهان نرمال WW افزایش نشان داده، تعداد 8 نقطه پروتیینی در گیاهان نرمال بیان نشده و 2 نقطه پروتیینی فقط در تیمار WD افزایش بیان داشت. تنش خشکی باعث تغییراتی در بیان پروتیینها گردید که اکثر تغییرات در ریشههای در معرض تنش خشکی رخ داد، از جمله افزایش بیانPR پروتیینها در پاسخ به خشکی افزایش و عدم بیان Heat shock پروتیینها و تیرودوکسین در ریشههای در معرض آب.

بررسی تنوع ژنتیکی برخی از جمعیتهای گیاه دارویی دم شیر در ایران با استفاده از نشانگر مولکولی RAPD
گیاه دمشیر با نام علمی Leonurus cardiaca از خانواده Lamiaceae (نعناعیان) و زیر خانواده Lamioideae تنها گونه موجود از جنس Leonurus در ایران است. وجود ترکیبات مختلفی چون فلاونوئیدها، ایریدوئیدها، تریترپنها، تاننها، استرولها، کاروتنوئیدها خواصی چون درمان بیماریهای قلب و معده و اختلالات عصبی را برای این گیاه به همراه دارد. در این تحقیق نشانگر مولکولی RAPD برای بررسی تنوع ژنتیکی ژرمپلاسم 47 نمونه گیاهی از 6 جمعیت وحشی جمعآوری شده گیاه دارویی دمشیر در ایران مورد استفاده قرار گرفت. تعداد 60 آغازگر تصادفی در انجام واکنش PCR روی DNA ژنومی استخراج شده از برگ آزمایش شد که 28 آغازگر نوارهای چند شکلی تولید نمودند و در کل 364 نوار DNA بدست آمد که تعداد 325 نوار چند شکل بودند. تجزیه کلاستر ژنوتیپها بر اساس ضریب تشابه دایس و به روش UPGMA انجام گرفت. میزان حداقل تشابه ژنتیکی در این تحقیق 08/0 بین نمونههای درگز 1 و ساری 5 و حداکثر آن بین نمونههای ساری 7 و ساری 3 معادل 87/0 بدست آمد. در تجزیه کلاستر ژنوتیپها در فاصله 20 دو کلاستر اصلی ایجاد شد که یکی از آنها شامل نمونههای درگز و کلاستر دیگر شامل نمونههای خوانسار، کرمان، سراب، طالقان و ساری بود. بررسی تنوع درون جمعیتها با استفاده از میانگین تنوع ژنتیکی نی و شاخص اطلاعاتی شانون نشان داد تنوع ژنتیکی در درون جمعیت سراب و طالقان (10/0= ,h 15/0= I) نسبت به سایر جمعیتها، بیشترین و در جمعیت ساری (05/0 = h ,08/0 = I) کمترین بوده است. تلاقی ژنوتیپهای جمعیت درگز با دیگر جمعیتها با توجه به فاصله ژنتیکی تعیین شده در این مطالعه میتواند گزینه مناسبی برای تولید هیبریدهایی با عملکردهای مورد نظر باشد.

مطالعه مورفولوژیکی و بیوشیمیایی جهت تشخیص تنوع ژنتیکی جمعیتهای یولاف زراعی
مطالعه صفات مورفولوژیکی و نشانگرهای مولکولی جهت تشخیص تنوع ژنتیکی گامی مهم و اساسی در اکثر برنامههای اصلاحی میباشد. در مطالعه حاضر از دادههای حاصل از صفات کمی و الگوی باندی پروتئینهای بذر برای ارزیابی تنوع ژنتیکی و طبقه بندی برخی از ژنوتیپهای یولاف زراعی استفاده گردید. برای این منظور 10 ژنوتیپ یولاف با آرایش طرح بلوکهای کامل تصادفی با سه تکرار مورد بررسی قرار گرفت. نتایج تجزیه واریانس دادهها نشان داد که ژنوتیپهای مورد بررسی از نظر برخی صفات مطالعه شده دارای اختلاف معنیدار هستند. تجزیهی خوشهای به روش UPGMA برای دادههای زراعی، ژنوتیپهای مورد ارزیابی را در سه گروه قرار داد. بیشترین سود ژنتیکی برای صفت طول برگ پرچم (74/94) مشاهده گردید. برای تعیین همبستگی بین صفات مورفولوژیکی و مولکولی از آزمون مانتل– هانزل (Mantel - Haenszel) استفاده گردید و همبستگی معنیداری بین صفات مورفولوژیکی و دادههای مولکولی مشاهده نشد. با توجه به فواصل ژنتیکی بین ژنوتیپها، در برنامههای بهنژادی جهت به دست آوردن مقدار هتروزیس، تلاقی ژنوتیپهای 212 با ژنوتیپهای 217، 202، 204 و 201 با توجه به ضرایب تشابه مورفولوژیکی قابل توجیه میباشد. در تحقیقات آتی، مطالعه آزمایشات مشابه به همراه تعداد بیشتری از ژنوتیپها توصیه میگردد.

جایگاه تاکسونومیکی جدایه های ایرانی ویروس موزائیک خیار از استان های کرمان و یزد
ویروس موزائیک خیار (CMV Cucumber mosaic virus,) یکی از ویروس های مهم کدوئیان در نقاط مختلف دنیاست. به منظور شناسایی و تعیین جایگاه تکاملی جدایههای این ویروس، تعدادی نمونه از استانهای کرمان و یزد جمعآوری گردید. آلودگی به CMV در این نمونهها توسط آزمون DAS-ELISA مورد بررسی قرار گرفت. بر اساس گیاه میزبان و مناطق نمونهبرداری، از نمونه های آلوده شش جدایه متفاوت، جهت مطالعات مولکولی انتخاب گردیدند. با استفاده از آزمون RT-PCR و آغازگرهای اختصاصی، یک قطعه 946 جفت بازی در برگیرنده ژن پروتئین پوششی، تکثیر، همسانه سازی و تعیین ترادف گردید. قطعات همسانه سازی شده جهت تعیین جایگاه تکاملی جدایههای ایرانی با ترادف های مشابه مربوط به سایر جدایه های CMV موجود در بانک ژن مقایسه شدند. آنالیز فیلوژنتیکی نشان داد که جدایههای بررسی شده در دو زیر گروه I و II قرار میگیرند که زیر گروه I نیز به دو زیر گروه IA وIB تقسیم میگردد. دو جدایه ایرانی CMV در زیر گروه IA و چهار جدایه دیگر در زیر گروه IB واقع گردیدند. بیشترین میزان تشابه ترادف نوکلئوتیدی در بین جدایههای ایرانی به میزان 1/99% دربین دو جدایه Ker.Ker.Pep و Ker.Ker.Mel.2 از زیر گروه IB و کمترین میزان تشابه به میزان 1/92% بین دو جدایه Ker.Jir.Cu از زیر گروه IA و Yaz.Yaz.Tom از زیر گروه IB تعیین گردید. جدایههای زیر گروه IB اختصاص به کشورهای شرق آسیا داشته و این بررسی اولین گزارش از وجود تعدادی ازجدایههای CMV مربوط به زیر گروه IB از ایران و خاورمیانه میباشد.

تهیه آزمایشگاهی وکتور T/A برای تسهیل همسانه سازی قطعات DNA حاصل از PCR
امروزه مهندسی ژنتیک جزء اجتناب ناپذیر پژوهشهای زیست شناسی نوین میباشد و همسانهسازی به عنوان یکی از اصلیترین بخشهای این علم، کاربرد گستردهای دارد. پلاسمیدها یکی از پرکاربردترین حاملها میباشند که امکان تکثیر ژن مورد نظر را به صورت نامحدود در میزبان عمدتا" باکتریایی فراهم میکنند. وکتورهای T/A نوعی از حاملهای پلاسمیدی میباشند که امکان همسانهسازی قطعه دیانای تکثیر شده با آنزیم Taq DNA polymerase (در واکنش زنجیرهای پلیمراز) را تسهیل می کنند. با توجه به محدودیت امکانات، خرید کیت همسانهسازی حاوی این پلاسمید ممکن است به سختی میسر باشد. به همین جهت تهیه این وکتور با کارایی خوب در آزمایشگاه حائز اهمیت است. در این پژوهش، بعد از تهیه سلول های مستعد (رقیب) از باکتری Escherchia coli، پلاسمید حلقوی و فاقد قطعه خارجی pTZ57R به این سلولها ترانسفورم گردید. پس از استخراج پلاسمید به روش لیزآلکالینی، با آنزیم EcoRV پلاسمید برش داده شده و خطی گردید. سپس آنزیم برشی غیرفعال گردید و نوکلئوتید تیمین به انتهای آزاد این پلاسمید خطی اضافه شد. سپس کارایی حامل تهیه شده طی واکنش اتصال دیانای به پلاسمید و متعاقب آن، ترانسفورماسیون باکتری تأیید گردید. بعلاوه، بخشی از ژنوم پوتیویروس که با جفت آغازگر یونیورسال مورد تکثیر قرار گرفته بود در پلاسمید ساخته شده مورد همسانه سازی وترادف یابی قرار گرفت که در مقایسه با توالیهای همقطار موجود در پایگاه ژن بانک بعنوان ویروس موزائیک سویا تشخیص داده شد.
