الگوی بیان ژن های OSBP، CAT و BZIP در ارقام حساس و متحمل به خشکی سویا با استفاده از PCR در زمان واقعی
در شرایط تنش خشکی، سیستم علامت دهنده موجب القای ژنهای مشخصی در مقابل اثرات زیانآور تنشهای محیطی میشود. ژن BZIP از فاکتورهای رونویسی در سیگنالرسانی تنشهای غیرزنده بوده و در تنظیم پاسخ به تنشهای مختلف در گیاهان نقش دارد. این ژن با ABA فعال شده و باعث بسته شدن روزنهها میشود. ژن OSBP دارای نقش کلیدی در سیگنالدهی برخی واکنشهای فیزیولوژیکی در پاسخ به تنش میباشد. کاتالاز (CAT) از سری آنزیمهای احیاکننده است که تبدیل پراکسید هیدروژن به آب و مولکول اکسیژن را کاتالیز و از سلول در برابر اثرات سمی پراکسید هیدروژن حمایت میکند. در این تحقیق دو رقم سویا Williams (متحمل) و L17 (حساس) در شرایط گلخانهای کشت شدند. تیمار تنش خشکی از مرحله دو برگی به مدت 7 روز اعمال شد. استخراج RNA کل از برگ و ریشه در دو سطح شاهد و تنش صورت گرفت و سپس ساخت cDNA جهت استفاده در Real Time PCR انجام شد. از ژن خانهدار 18SrRNA برای نرمال کردن دادهها استفاده شد. تجزیه دادهها با استفاده از منحنیهای Ct نشان دهنده افزایش بیان ژن BZIP در تنش خشکی بود و این افزایش در رقم متحمل Williams حدود دو برابر رقم حساس L17 و در ریشه بیشتر از برگ بود. اختلاف بیان ژن OSBP تحت تنش خشکی بین دو رقم Williams و L17 معنیدار نگردید. بیان ژن کاتالاز در تنش خشکی در رقم Williams حدود دو برابر رقم L17 و همچنین در ریشه بیشتر از برگ بود. با توجه به اینکه افزایش بیان این ژنها منجر به افزایش تحمل خشکی میشود، انتقال این ژنها در راستای افزایش تحمل به خشکی در ژنوتیپهای پرعملکرد سویا مناسب و موثر ارزیابی گردید.

مطالعه اثر ناحیه ژنومی حامل ژن Hd1 بر زمان خوشه دهی در برنج
عبور از مرحله رویشی به مرحله زایشی موجب تنظیم زمان خوشهدهی در برنج میگردد که از صفات مرتبط با سازگاری این گیاه به نواحی کاشت مختلف یا فصول متفاوت، میباشد. چندین ژن مرتبط با زمان خوشهدهی در برنج شناسایی شده است که یکی از مهمترین آنها ژن Hd1 بر روی کروموزوم شماره 6 برنج میباشد. در این مطالعه دو رقم برنج شامل ارقام صدری (به عنوان والد بخشنده زودرس) و ندا (به عنوان والد دورهای دیررس)، به منظور بررسی اثر ناحیه ژنومی حامل ژن Hd1 بر زمان خوشهدهی در برنج، تلاقی داده شدند و با دو بار تلاقی برگشتی نسل BC2 ایجاد گردید. سپس یک بوته هتروزیگوت از نسل BC2 خودگشن گردید تا جمعیت BC2F2 جهت انجام مطالعات فنوتیپی و مولکولی به دست آید. جمعیت مورد مطالعه از نظر زمان خوشهدهی دارای توزیع فنوتیپی پیوسته بوده و برخی افراد جمعیت تفکیک متجاوز نسبت به والد دیررس نشان دادند. بر اساس ناحیه InDel موجود در اگزون شماره یک ژن Hd1 یک نشانگر کارکردی اختصاصی طراحی شد که تفاوت باندی واضحی بین والدین تلاقی ایجاد نمود. از اینرو، این نشانگر جهت ارزیابی الگوی آللی Hd1 در جمعیت BC2F2 استفاده گردید. بررسی نحوه تفکیک آللی نشان داد که مکان ژنی Hd1 در جمعیت مورد مطالعه از الگوی تفرق مورد انتظار مندلی (با نسبت 1:2:1) پیروی نمود. نقشهیابی فاصلهای مرکب (CIM) با نشانگرهای ریزماهواره نشان داد که ناحیه ژنومی در حد فاصل نشانگرهای Hd1-RM527 ارتباط زیادی (5/7<LOD) با زمان خوشهدهی داشت و حدود 28 درصد از تغییرات فنوتیپی را توجیه نمود. اثر افزایشی آلل والد بخشنده، منفی و در حدود 4/3- روز برآورد گردید. نتایج این بررسی بیانگر اثر معنیدار ژن Hd1 بر زمان خوشهدهی در برنج میباشد و از نشانگر کارکردی توسعه یافته در این تحقیق میتوان برای ردیابی ژن Hd1 در جمعیتهای در حال تفرق و گزینش به کمک نشانگر (MAS) برای زودرسی استفاده کرد.

ارزیابی تنوع ژنتیکی و گروه بندی لاین های پیشرفته آفتابگردان (Helianthus annus L.) با استفاده از نشانگرهای ISSR
آفتابگردان از جمله مهمترین محصولات دانه روغنی می باشد. بررسی تنوع ژنتیکی و گروه بندی ژنوتیپ ها و لاین ها از مهمترین عوامل پیشبرد برنامههای به نژادی گیاهی است. در این مطالعه، تنوع ژنتیکی و گروهبندی تعدادی از لاین های پیشرفته اصلاحی آفتابگردان حاصل از برنامه های اصلاحی مختلف، با استفاده از نشانگرهای ISSR انجام شد. تعداد 15 آغازگر ISSR از میان 21 آغازگر با تکثیر مناسب در واکنش زنجیره ای پلی مراز برای انگشت نگاری لاین های آفتابگردان استفاده شدند. در مجموع تعداد 70 نوار بوسیله آغازگرهای ISSR تکثیر شد که تعداد 27 نوار در بین تمام لاینها یک شکل و 43 نوار چندشکل بودند. نتایج نشان داد که بیشترین مقدار محتوای اطلاعات چندشکل مربوط به آغازگر 807 UBC (4/0) و کمترین مقدار آن مربوط به آغازگر 804 UBC (15/0) می باشد. با استفاده از ضریب تشابه دایس، کمترین شباهت (65/0) بین لاین های SF25 با SF278 و بیشترین شباهت (93/0) بین دو لاین HA336 و SF315 مشاهده شد. تجزیه خوشه ای بر اساس الگوریتم UPGMA لاینهای مورد مطالعه را در 8 گروه قرار داد. با توجه به نتایج تجزیه به مختصات اصلی، هر مولفه درصد کمی از تغییرات را توجیه می نمود که این امر نشان دهنده پراکنش ژنومی مناسب آغازگرهای ISSR در این تحقیق بود.

تجزیه ارتباط صفات مورفولوژیک در انگور با استفاده از نشانگرهای SSR و AFLP
انگور متعلق به تیره Vitaceae است و به علت داشتن مصارف متعدد به عنوان یک منبع مهم غذایی همیشه مورد توجه بوده است. به منظور شناسایی نشانگرهای مرتبط با 14 صفت شامل حجم آب میوه، درصد جوانهزنی دانه گرده، درصد تشکیل میوه، میانگین وزن تک خوشه و وزن تک حبه، وزن گوشت میوه، وزن تک بذر، طول و عرض خوشه و سیستم باردهی، مواد جامد محلول میوه، مقدار اسید و pH میوه در 48 رقم انگور ایرانی موجود در کلکسیون مرکز تحقیقات کشاورزی و منابع طبیعی استان آذربایجان غربی از 22 جفت آغازگر SSR و 7 ترکیب آغازگری AFLP استفاده گردید. تجزیه رگرسیون گام به گام رابطه معنیداری بین تعدادی از نشانگرهای SSR و تمامی 14 صفت مورد مطالعه در ارقام انگور نشان داد. در کل 49 آلل از 18 نشانگر SSR ارتباط معنیدار با تغییرات 14 صفت نشان دادند. وزن حبه و وزن گوشت میوه هرکدام با 7 آلل بیشترین و صفات pH میوه، اسید میوه، حجم آب میوه و درصد جوانه زنی گرده با یک آلل کمترین تعداد آلل مرتبط را داشتند. از 7 جفت ترکیب آغازگری AFLP مورد استفاده، 49 قطعه رابطه معنیداری با تغییرات 14 صفت نشان دادند. تعدادی از این قطعات برای صفات مختلف مشترک بودند. وزن گوشت میوه با 11 نشانگر مرتبط بیشترین و صفات مواد جامد محلول، درصد تشکیل میوه، جوانه زنی گرده و عرض خوشه با یک نشانگر مرتبط، کمترین تعداد نشانگرهای مرتبط AFLP را به خود اختصاص دادند.

تاثیر تغییرات تک نوکلئوتیدی در ژنهای کاپاکازئین، استئوپونتین وPPARGC1α روی صفات تولید شیر و کیفیت تولید پنیر نژاد براون سوئیس
شیر از دیرباز به عنوان غذای کامل و تامین کننده بخشی از نیازهای تغذیهای روزانه انسان مورد توجه بوده است. یکی از ترکیبات مهم شیر پروتئینها میباشند که در صنایع فرآوری شیر و تکنولوژی تولید محصولات شیری مانند پنیرسازی، فاکتور مهمی بشمار میآیند. عوامل متعددی از جمله ژنتیک میتوانند بر مقدار و ترکیبات شیر و نهایتا کمیت و کیفیت پنیر تاثیر گذارند. در این راستا، از طریق تکنیکهای ژنتیک مولکولی نواحی متعددی روی ژنوم گاو از جمله کروموزوم 6 شناسایی شده است. در این پژوهش تاثیر 3 ژن کاپاکازئین، اوستئوپونتین و PPARGC1α از کروموزوم فوق روی صفات تولیدی شیر و راندمان تولید پنیر بررسی شد. استخراج DNA به روش استخراج نمکی از نمونه خون 100 راس گاو براون سوئیس انجام شد. ژنوتیپها به روش PCR-RFLP و با استفاده از آنزیمهای HinfI، BsurI،NheI و BsrI تعیین شدند. ارزشهای اصلاحی با نرم افزار ِDFREML برآورد و چهار فاکتور راندمان، درصد چربی و پروتئین و ماده خشک پنیر اندازهگیری شدند. همچنین، ارتباط چندشکلیها با ارزش اصلاحی صفات و کیفیت محصول پنیر در سطح معنیداری 5% با رویه GLM بررسی گردید. فراوانیهای ژنی و ژنوتیپی محاسبه شد و نتایج نشان داد که بجز کاپاکازئین بقیه جایگاهها در حالت تعادل هاردی-واینبرگ قرار داشتند. در این تحقیق ژنوتیپ روی محصول پنیر اثر معنیدار نداشت اما تاثیر همزمان ژنوتیپ PPARGC1α -T19C و کاپاکازئین روی ارزشهای اصلاحی تولید شیر معنیدار بود. ارتباط معنیدار بین چندشکلیهای تک نوکلئوتیدی با صفات تولید شیر، راهکار مناسبی جهت بهبود ارزیابیهای گاو شیری است. استفاده از روشهای شناسایی تک نوکلئوتیدهای متراکم در پژوهشهای تکمیلی، توصیه میشود

توالی یابی و تجزیه و تحلیل اطلاعات بدست آمده یکی از بهترین و رایج ترین روشها در مطالعات تنوع ژنتیکی می باشد. ژن PIT-1 به نام فاکتور 1 ژن هورمون رشد معروف است. این ژن عضوی از خانواده فاکتورهای نسخه برداری است و در طیور روی کروموزوم 1 با طول تقریباً kb14 قرار دارد و دارای 7 اگزون و 5 اینترون می باشد. هدف از انجام این پژوهش تجزیه و تحلیل توالی ناحیه اینترون 5 ژن PIT-1 در مرغان گوشتی لاین آرین و ترسیم درخت فیلوژنی آن با سایر نژادهای مرغ بود. برای انجام این پژوهش از 7 قطعه مرغ از هر دو جنس مرغان گوشتی لاین آرین نمونه خون جمع آوری شد . پس از استخراج DNA، ژن مورد نظر توسط آغازگرهای اختصاصی تکثیر و قطعات تکثیر شده پس از خالص سازی به صورت رفت و برگشت توالی یابی شدند. تعداد 5 هاپلوتیپ مختلف بر اساس 6 نوکلئوتید چند شکل موجود در توالی ها تعیین گردید و توالی های نهایی بدست آمده از هر هاپلوتیپ با طول تقریبی260 جفت باز که شامل 92/21 درصد آدنین، 08/18 درصد گوانین، 69/32 درصد سیتوزین،31/27 درصد تیمین بود. پس از اطمینان از صحت توالی یابی در پایگاه اطلاعاتی بانک ژن با شماره های دسترسیJQ946630- JQ946636 ثبت شدند. پس از اخذ توالی های مشابه ژن PIT-1دیگر نژادهای موجود در بانک جهانی ژن درخت فیلوژنی با استفاده از آنها ترسیم شد. نتایج فیلوژنی مشخص کرد که مرغ آرین ایران با مرغان آمریکایی در یک گروه قرار دارند.

بررسی امکان ریزازدیادی گیاه زینتی دراسنا با استفاده از روش کشت درون شیشهای
دراسنا (Dracaena) از جمله گیاهان ارزشمند و زینتی در ایران و جهان محسوب شده که تکثیر انبوه و ایجاد تنوع ژنتیکی در آن در تحقیقات ریز ازدیادی و بهنژادی از اهمیت ویژهای برخوردار است. در این تحقیق بهمنظور کالوسزایی و باززایی رقم Tricolor از گونه Dracaena marginata، از قطعات کوچک جوان ساقه با طول cm 5/0 استفاده شد. بهمنظور بررسی کنترل آلودگیهای سطحی بافتهای فوق از هیپوکلریت سدیم (غلظتهای 1%٬ 25/1% 5/1%) در زمانهای 15 و 10 دقیقه استفاده گردید. غلظتهای متفاوت از تنظیم کنندههای رشد 2,4-D، NAA و Kinetin در محیط پایه MS جهت ارائه مناسبترین محیط غذایی بهمنظور القاء کالوس، باززایی شاخساره از کالوس و ریشهزایی شاخساره مورد بررسی قرار گرفت. جهت انجام بخشهای مختلف این پژوهش از طرحهای آزمایشی مناسب استفاده گردید. نتایج بدست آمده نشان میدهد که مناسبترین تیمار برای ضدعفونی قطعات فوق همراه با بالاترین درصد سلامت بافتها مربوط به غلظت 25/1% هیپوکلریتسدیم به مدت 15 دقیقه بود. مناسبترین غلظتهای تنظیم کنندههای رشدی در محیط MS جهت القاء کالوس در قطعات کوچک ساقه دراسنا شامل mgL-1 5/0 2,4-D و یا mgL-1 1 NAA و میزان mgL-1 1 از Kinetin بهمنظور باززایی شاخساره از کالوس و غلظت mgL-11 از NAA بهمنظور القاء ریشهزایی در شاخساره در محیط پایه MS تعیین شد. با توجه به تعیین محیط غذایی مناسب جهت القاء کالوس در بافت ساقه دراسنا و امکان باززائی شاخساره در این تحقیق، امکان بهنژادی و انتقال ژنهای مطلوب به این گیاه زینتی ارزشمند تسهیل شده است

تشخیص تعدادی از عوامل باکتریایی سقط جنین در گوسفند افشاری با استفاده ازReal Time PCR و تعیین حساسیت آغازگر مربوط به کمپیلوباکتر
ارایه روشی مناسب برای تشخیص سریع و دقیق باکتریهای مولد بیماری سقط جنین از اولویتهای کنترل و درمان به موقع این بیماری است. هدف از این تحقیق بهینه کردن یک روش مبتنی بر Real Time PCR به صورت همزمان برای چهار باکتری مهم مولد سقط جنین (کمپیلوباکتر، بروسلا، یرسینیا و سالمونلا) بود. تعداد 217 نمونه سواب واژینال (132 نمونه از دامهای سقط کرده و 85 نمونه از دامهایی که برههای سالم به دنیا آوردند) از گوسفندهای استان زنجان گرفته شده و برای سه باکتری سالمونلا، یرسینیا و بروسلا نیز نمونههای خالص آنها از مرکز جمعآوری قارچها و باکتریهای صنعتی ایران PTCC خریداری شد. پس از استخراج DNA، آغازگر های اختصاصی بروسلا، سالمونلا و یرسینیا بر روی نمونه DNA باکتریهای خریداری شده مورد بررسی قرار گرفتند و پس از مشخص شدن کمپیلوباکتر به عنوان عامل سقط جنین در تعدادی از نمونههای استان زنجان (68 نمونه مثبت از 132 دام سقط کرده و 29 نمونه از 85 دام سالم)، دقت آغازگر کمپیلوباکتر در رقتهای مختلف با استفاده از Real Time PCR نیز سنجیده شد. نتایج این بررسی نشان داد که همه باکتریها در شرایط یکسان با آغازگرهای اختصاصی خود به صورت همزمان با Real Time PCR قابل تکثیر بوده و آغازگر کمپیلوباکتر نیز در تمام رقتهای ایجاد شده حتی تا یک کپی از ژنوم هم قادر به تشخیص و تکثیر باکتری بوده است.

بررسی اثر مسمومیت زایی نانو ذره نقره بر سیستمهای زیستی و اکولوژیکی
نانونقره بهدلیل خواص ضد باکتریایی، بهطور گستردهای در بسیاری از محصولات شامل: لباس، رنگها، پلاستیکها، ظروف نگهداری غذا، باندهای زخم، لوازم بهداشتی و وسایل و لوازم خانگی از قبیل یخچال و ماشین لباسشویی استفاده شده است. اما بهدلیل پایین بودن قیمت آنتیبیوتیکها، نانونقره در تغذیه دام و طیور بهعنوان محرک رشد کمتر استفاده شدهاست. اخیرا با توجه به ممنوعیت استفاده از آنتیبیوتیکهای محرک رشد در تغذیه دام و طیور استفاده از مواد جایگزین مانند ترکیبات نانونقره مورد توجه قرار گرفتهاست. نتایج آزمایشات مختلف روی طیور نشان داده است که ترکیبات نانو نقره اثر منفی بر رشد و توسعه این گونه نداشتهاست. نتیجه آزمایش ما در دانشگاه شهید باهنر کرمان که روی جوجه های گوشتی در فازهای مختلف تولیدی انجام شد نشان داد که استفاده از سطوح مختلف نانو نقره (ppm 5/1 و 1و 5/0) در آب آشامیدنی طیور اثر منفی بر عملکرد جوجه ها نداشت ولی بطور معنیداری تعداد باکتریهای مفید روده یعنی باکتریهای اسید لاکتیک را افزایش داد. با این حال نگرانیهایی در رابطه با استفاده این مواد بعنوان مواد ضد باکتریایی وجود دارد. زیرا این مواد در بافت های مختلف حیوان ذخیره شده و پتانسیل مسمومیت زایی برای حیوان و انسان را داشته و همچنین ممکن است باعث آلودگی محیط زیست هم بشوند. علیرغم توسعه تجارتی استفاده از نانو نقره در محصولات مختلف اطلاعات اندکی در رابطه با اثرات زیست محیطی این مواد وجود دارد. زیرا که مقادیر اندک این مواد در حد نانو گرم در لیتر میتواند زندگی پروکاریوتها، بیمهرگان و ماهیها را تحت تاثیر قرار داده و باعث آلودگی محیط زیست هم بشود. با توجه به اینکه سازوکارهای مسمومیتزایی این مواد هنوز بخوبی روشن نیست، مطالعه حاضر نتایج حاصل از تحقیقاتی که در زمینه مسمومیتزایی نانوذرات نقره در گونههای مختلف جانوری انجام شده را بررسی و پیشنهادات و راه حلهایی را برای درک بیشتر نقش نانو نقره در مسمومیت زایی و آلودگی های زیست محیطی ارائه میکند.

همسانه سازی فرم اصلاح شده ژن tPAانسانی در ناقل کلروپلاستی و باززایی گیاهان ترانس پلاستومیک توتون
گیاهان جایگزین مناسبی برای سیستمهای معمول بیان پروتئینهای نوترکیب و داروهای زیستی، نظیر حیوانات یا سیستمهای میکروبی تراریخته میباشند. به دلیل پلیپلوئیدی بالای ژنوم پلاستید گیاهی، در صورت تراریختی کلروپلاست، هزاران نسخه از ژن خارجی در هر سلول وارد و حجم بسیار بالایی از پروتئین نوترکیب تولید میشود. پس از سرطانها، بیماریهای قلبی- عروقی دومین عامل مرگ و میر انسانی هستند. فعالکننده پلاسمینوژن بافتی انسانی (tPA) یکی از مهمترین پروتئینهای نوترکیب داروئی است که جهت از بین بردن لخته خون در قسمتهای مختلف بدن از جمله رگهای خونی قلب و مغز استفاده میشود. فرم اصلاح شده پلاسمینوژن بافتی (K2S) دارای نیمه عمر پلاسمایی طولانیتر، قدرت نفوذ در لخته و فعالیت فیبرینولیتیک بالاتر میباشد. در این مطالعه، بهمنظور انتقال ژن K2S به کلروپلاست گیاه توتون، پس از طراحی سازه، ژن هدف در ناقل کلروپلاستی pKCZ درج و سپس در باکتریcoli .E همسانهسازی گردید. پس از شلیک موفق ناقل حاوی ژن K2S (pKCZK2S) به ریزنمونههای برگی با استفاده از تکنیک بیولستیک، ریزنمونهها بر روی محیط انتخابی حاوی 500 میلی گرم در لیتر آنتی بیوتیک اسپکتینومایسین قرار گرفتند. پس از باززایی نوساقههای رشد یافته بر روی محیط انتخابی، به منظور رسیدن به هموپلاسمی چهار دوره انتخاب و باززایی در حضور آنتیبیوتیک اسپکتینومایسین انجام شد. حضور، درج در جایگاه مشخص، بیان ژن K2S و هموپلاستی در گیاهان ترانسپلاستومیک با روشهای PCR، RT-PCR و سادرن بلات به تایید رسید.

مطالعه سیتوژنتیک و بهینه سازی روش دورگ سازی ژنومی در محل در جنس پسته (Pistacia spp.)
پسته (Pistacia vera) یکی از مهمترین محصولات باغی شناخته شده ایران در دنیا میباشد که نام ایران را با داشتن بیشترین ارقام تجاری دنیا به عنوان مهمترین مبنع ژرم پلاسم این گیاه مطرح نموده است. با این وجود تعداد مطالعات کروموزومی روی این محصول محدود و گزارشات موجود حاکی از تناقضاتی در تعداد کروموزومها میباشد. در این تحقیق گونههای پسته موجود در ایران شامل ارقام اهلی و گونههای غیر اهلی خنجوک و بنه با استفاده از روشهای کلاسیک رنگآمیزی کروموزوم و روش دورگ سازی ژنومی در محل مورد مطالعه قرار گرفتند. دو روش استو-آهن هماتوکسیلین و فولجن برای رنگآمیزی کروموزومها به کار گرفته شد. بر اساس نتایج، رنگآمیزی با فولجن، روش مناسبتری برای رنگآمیزی کروموزومهای پسته در مقایسه با روش دیگر بود. تعداد کروموزمها در ارقام اوحدی، اکبری، احمدآقایی، نیش کلاغی و خنجری دامغان 15 جفت (30=n2) در حالیکه ارقام ایتالیایی و قزوینی، واریته وحشی سرخس و گونههای خنجوک و بنه دارای 14 جفت کروموزوم (28=n2) بودند. در آزمایش دورگ سازی ژنومی در محل، تابش فلورسنتی که دال بر هیبریداسیون پروبهای ژنومی خنجوک و بنه با نمونههای کروموزومی ارقام اهلی باشد، مشاهده نگردید. عدم مشاهده هیبریداسیون قابل ردیابی میتواند ناشی از مشتق شدن این کروموزومها از ردههای مختلف باشد.

تجزیه و تحلیل ژنتیکی و فیلوژنتیکی ناحیه کنترل ژنوم میتوکندریایی مرغ های بومی خراسان
مطالعه بر روی مرغهای بومی می تواند به حفظ تنوع ژنتیکی جمعیتها، پیاده نمودن برنامههای اصلاح نژادی و بدست آوردن اطلاعات در رابطه با ساختار ژنتیکی آنها کمک کند. توالییابی ژنوم میتوکندری یکی از کاربردی ترین روشها برای تعیین رابطه فیلوژنتیکی بین جمعیتها و گونههای نزدیک به هم محسوب میشود. هدف از این مطالعه بررسی توالی نوکلئوتیدی ناحیه کنترل از DNA میتوکندریایی مرغهای بومی خراسان و مطالعات ژنتیکی و فیلوژنتیکی بود. در این مطالعه از 5 قطعه مرغ بومی خراسان نمونه خون تهیه شد پس از استخراج DNA از نمونههای خون، تکثیر ناحیه کنترل به طول 845 جفت باز با آغازگرهای اختصاصی صورت گرفت. توالییابی ناحیه تکثیر شده به روش سانگر انجام گردید. با استفاده ازتوالیهای مشابه ژنوم میتوکندریایی دیگر نژادهای مرغ موجود در پایگاه NCBI، درخت فیلوژنتیکی ترسیم شد و ماتریس فواصل ژنتیکی بین مرغ بومی خراسان با دیگر نژادها برای ناحیه کنترل ژنوم میتوکندریایی تشکیل گردید. نتایج نشان داد که در بین توالیهای نمونههای مورد مطالعه، تفاوتهای هاپلوتایپی وجود نداشت. نتایج آزمون فیلوژنتیکی نشان داد حداقل فاصله ژنتیکی بین مرغهای بومی خراسان با مرغهای مرندی از ایران، لگهورن سفید، Buff Cochin و satsuma dori از ژاپن، Lv erwu از چین، Denizli و Gerze از ترکیه، Dandarawi از مصر و هند وجود دارد. بنابراین، میتوان نتیجه گرفت که قرابت زیادی بین مرغهای بومی خراسان از ایران با مرغهای آسیایی وجود دارد.
