آنالیز مولکولی و فیزیولوژیکی پیری برگ پرچم و انتقال مجدد مواد فتوسنتزی در گندم نان تحت تنش خشکی انتهایی
در مطالعه فیزیولوژیکی انتقال مجدد مواد فتوسنتزی از ساقه به دانه در طی پر شدن دانهها مواد ژنتیکی جهشیافته ابزاری ارزشمند به شمار میآیند. دو لاین جهشیافته پیشرفته گندم نان (T-67-60 و T-65-7-1) به همراه رقم تیپ وحشی آنها (رقم طبسی) در دو شرایط رطوبتی (مطلوب و 40-30 درصـد ظرفیت مزرعه) به صورت یک آزمایش فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار کشت شدند. اعمال تنش در مرحله ظهور کامل سنبله (زادوکس 60) آغاز شد و نمونهبرداریها در 5 مرحله (0، 7، 14، 21 و 28 روز پس ازگردهافشانی) انجام شد. بر اساس نتایج انتقال مجدد ساقه و کارایی آن تحت تنش خشکی انتهایی در لاینهای جهشیافته به طور معنیداری بیشتر از رقم تیپ وحشی بود. علت این امر قدرت مخزن بالاتر و بروز پیری زودرس در لاینهای جهشیافته نسبت به رقم تیپ وحشی بود. با توجه به روند تغییرات محتوای کلروفیل و بیان نسبی ژنهای کلیدی فتوسنتزی روبیسکو (زیرواحدهای کوچک و بزرگ روبیسکو و روبیسکو اکتیواز) در ژنوتیپها طی پر شدن دانهها به نظر میرسد تجزیه کلروپلاست و سیستم فتوسنتزی در اثر پیری القاء شده توسط تنش خشکی در لاینهای جهشیافته با شدت بیشتری نسبت به رقم تیپ وحشی اتفاق افتاده است. از آنجایی که پدیده پیری تحریککننده انتقال مجدد مواد فتوسنتزی از ساقه به دانه است، انتقال مجدد ساقه و کارایی آن در لاینهای جهشیافته بیشتر از رقم تیپ وحشی بود. رقم تیپ وحشی نیز بیشتر با تداوم فتوسنتز جاری برگها در جهت پر کردن دانهها عمل کرده است.

بررسی رشد، تکثیر و ریشهزایی ارقام مختلف گلابی بومی ایران به منظور حفاظت درون شیشهای
تکثیر رویشی گلابی از طریق روشهای معمول قلمه چوب نرم یا سخت مشکل میباشد. بنابراین امکان تکثیر و ایجاد کلکسیون درون شیشهای، در 10 رقم بومی گلابی ایران مورد مطالعه قرار گرفت. برای این منظور ابتدا صفات رشد گیاهچهها از قلمههای تکگره نرم این ارقام بر روی محیط کشت MS حاوی تنظیم کنندههای رشد بررسی شد. در این مرحله پس از گذشت سه هفته از کشت ریز نمونهها، ارقام نطنزی و قوسی با میانگین 75/4 روز زودترین زمان القاء رشد و رقم خوج با میانگین 75/20 روز دیرترین زمان القاء رشد را نشان داد. ارقام قوسی و نطنزی بیشترین تعداد برگ و ارتفاع شاخساره را به خود اختصاص دادند. سپس تکثیر درون شیشهای گیاهچههای این ارقام در محیط کشت QL تغییر یافته حاوی تنظیم کنندههای رشد مورد مطالعه قرار گرفت. در گیاهچههای تکثیر شده درون شیشهای، رقم قوسی به صورت متوسط بیشترین تعداد برگ تکثیر شده و بالاترین ارتفاع گیاه را تولید کرد. ارقام بیروتی و شاه میوه کمترین رشد را نشان دادند. همچنین ریشهزایی ارقام منتخب گلابی در غلظتهای مختلف اکسین (IBA) بر روی محیط کشت QL تغییر یافته مطالعه شد. در غلظتهای 5/0 و 2 میلیگرم در لیتر IBA در هیچ یک از ارقام ریشهزایی مشاهده نشد، در حالیکه همه ارقام مورد بررسی در غلظت 1 میلیگرم در لیتر ریشهدار شدند. بیشترین تعداد ریشه در رقم قوسی و کمترین میزان آن در ارقام نطنزی و سردرودی مشاهده گردید. رقم شاه میوه دارای بیشترین طول ریشه و ارقام سردرودی و قوسی دارای کمترین طول ریشه بودند. نتایج این تحقیق نشان داد که تکثیر و ریشهدار نمودن ارقام گلابی بومی ایران در شرایط درون شیشه به راحتی امکانپذیر بوده ولی صفات رشد و تکثیر آنها تحت تاثیر ژنوتیپ (رقم) میباشد. همچنین در این تحقیق اولین کلکسیون حفاظت درون شیشهای ذخایر ژنتیکی گلابی در ایران با موفقیت ایجاد گردید.

نقشهیابی صفات سنبله و دانه با استفاده از خانوادههای F3 و F4 حاصل از تلاقی بیچر× کویر در جو
جو پس از ذرت، گندم و برنج یکی از مهمترین غلات جهان میباشد. بهمنظور نقشهیابی صفات سنبله و دانه در دو نسل F3 و F4 حاصل از تلاقی ارقام بیچر× کویر جمعیتی شامل 103 خانواده F3 و F4 جو در قالب طرح بلوکهای کامل تصادفی طی دو سال زراعی 95-1393 در سه تکرار کشت شد. برای صفات مورد ارزیابی تفکیک متجاوز مشاهده شد که نشاندهنده وجود ترکیبات آللی متفاوت در والدین بود. نقشه پیوستگی با استفاده از نشانگرهای SSR، iPBS،IRAP و ISSR تهیه شد و 640 سانتی مورگان از ژنوم جو را پوشش داد. فاصلهی بین دو نشانگر 69/9 سانتی مورگان برآورد گردید. مکانیابی فاصلهای مرکب پانزده QTL در دو نسل مکانیابی نمود. برای صفات طول ریشک، قطر دانه و طول دانه در هر دو نسل F3 و F4 QTLهایی شناسایی شد. برای طول دانه و تعداد سنبلچه در نسل F4 به ترتیب QTLهای بزرگ اثر با ضریب تبیین 2/12 و 15، با درصد LOD 62/3 و 9/2 در فاصلهی 5/3 و 5/2 سانتیمورگان از نشانگر شناسایی شدند. صفات طول دانه و قطر دانه در نسل F3 با نشانگر iPBS2076-5 پیوسته و هممکان بودند. برای صفات طول ریشک، تعداد سنبلچه، طول دانه و قطر دانه نیز هممکانی شناسایی شد. شناسایی QTLهای کنترلکننده صفات میتواند در بهبود عملکرد مؤثر باشد. انتظار میرود بتوان پس از تعیین اعتبار در مکانها و جمعیتهای مختلف از QTLهای شناسایی شده در این بررسی در برنامههای انتخاب به کمک نشانگر و مکانیابی دقیق استفاده نمود.

بررسی چند شکلی اگزون 17 ژن DGAT1 و ارتباط آن با درصد چربی شیر گوسفندان نژاد کردی ایستگاه اصلاح نژاد شیروان
دیآسیلگلیسرول آسیلترانسفراز 1 (DGAT1) یک آنزیم میکروزومی است که مرحله نهایی سنتز تریگلیسریدها یعنی تبدیل دی آسیل گلیسرول به تری آسیل گلیسرول را تسریع میکند. هدف از این پژوهش، مطالعه چندشکلی اگزون 17 ژن DGTA1 و بررسی ارتباط آن با رکوردهای درصد چربی شیر گوسفندان نژاد کردی ایستگاه اصلاحنژاد حسینآباد شیروان بود. بدین منظور از 100 رأس میش بطور تصادفی رکورد شیر گرفته شد و با دستگاه اکومیلک توتال، درصد چربی آن تعیین گردید. همچنین، DNA این میشها استخراج و با استفاده از یک جفت آغازگر اختصاصی، یک قطعهی 309 جفت بازی اگزون 17 این ژن تکثیر شد. ژنوتیپها به روش PCR-RFLP و هضم با آنزیم اندونوکلئاز ALuI تعیین شدند. فراوانی آلل C و T این ژن به ترتیب 65/0 و 35/0 محاسبه شد. آنالیز آماری با رویه GLM نرمافزار SAS 9.1 انجام شد و بیانگر ارتباط معنیدار ژنوتیپها با درصد چربی شیر میشها بود (05/0PDGTA1 میتوان برای افزایش درصد چربی شیر گوسفند در برنامههای انتخاب به کمک نشانگر استفاده کرد.

شناسایی جایگاههای ژنی مؤثر بر عدم تقارن اسکلتی در بلدرچین ژاپنی
عدم تقارن که نشاندهنده استرس ژنتیکی یا محیطی در موجودات است، میتواند در ناپایداری تکاملی حیوانات نقش داشته باشد. ثابت شده است که این عدم تقارن میتواند عامل رشد کندتر، باروری کمتر و بقای محدودتر موجودات باشد. مطالعه حاضر بهمنظور شناسایی جایگاههای ژنی مؤثر بر این عدم تقارن در استخوان متاتارسوس بلدرچین ژاپنی صورت گرفت. بدین منظور از یک طرح سه نسلی F2 حاصل از تلاقی متقابل دوسویه متفاوت بلدرچین ژاپنی (تخمگذار یا سفید و گوشتی یا وحشی) و تشکیل نسل دوم استفاده گردید. با تلاقی 34 پرنده از نسل دوم با یکدیگر، 422 پرنده نسل سوم ایجاد شد. رکوردهای فنوتیپی مربوط به وزن و طول استخوانهای مربوط به 403 پرنده ثبت شدند. 414 قطعه از پرندگان هر سه نسل (383 قطعه از نسل سوم) برای هشت نشانگر ریزماهواره تعیین ژنوتیپ شدند. دادههای حاصل با استفاده از روش نقشه یابی درون فاصلهای آنالیز شده و QTL مربوط به 12 صفت شناسایی شد. در این تحقیق، شش جایگاه ژنی در فواصل 59، 70، 134، 156، 164 و 184 سانتی مورگان روی کروموزوم شماره یک شناسایی شد. واریانس ناشی از جایگاههای شناسایی شده، 37/0 تا 29/5 درصد از واریانس فنوتیپی صفات موردنظر را شامل میشد.

ارزیابی فعالیت برخی از آنزیمهای دفاعی نخود (Cicer arietinum L.) تحت تنش سرما
گیاهان جهت بقا و تحمل تنشهای محیطی نیازمند مکانیسمهای دفاعی متعددی هستند. در این پژوهش فعالیت آنزیمهای اکسیداز متناوب (AOX) میتوکندری، سوپراکسید دیسموتاز (SOD) و کاتالاز (CAT) ، میزان نشت الکترولیتی (ELI) و پراکسید هیدروژن (H2O2) به عنوان شاخصهای خسارت سلولی در دو ژنوتیپ حساس (ILC533) و متحمل (Sel 96TH11439) نخود زراعی (Cicer arietinum L.) تحت تنش سرمای 4 درجه سانتیگراد ارزیابی شد. نتایج تجزیه آماری نشان داد که اختلاف معنیداری بین ژنوتیپها تحت تیمارهای دمایی وجود داشت. تحت تنش سرما به موازات کاهش شاخصهای خسارت، فعالیت آنزیم AOX افزایش معنیداری یافت. بیشترین فعالیت این آنزیم در ژنوتیپ متحمل در روز ششم پس از تنش سرما مشاهده شد در حالیکه تحت این شرایط میزان فعالیت این آنزیم در ژنوتیپ حساس بهطور معنیداری کمتر از ژنوتیپ متحمل بود. افزایش فعالیت آنزیم AOX که با خسارت سلولی کمتر (نتایج ELI و H2O2) بهخصوص در روز ششم پس از تنش سرما همراه بود، بیانگر اهمیت این آنزیم در تحمل به سرما در گیاه نخود میباشد. افزایش معنیدار و همزمان الگوی فعالیت آنزیمهای SOD و CAT ضمن تایید نتایج فعالیت AOX، درجه تحمل ژنتیکی نخود به سرما را افزایش داده و یا اینکه سبب بهبود گیاه پس از اعمال تنش میشود. چنین شاخصهایی ممکن است در ارزیابی ژنوتیپهای نخود تحت تنش سرما و یا بکارگیری آنها در برنامههای اصلاحی مفید باشند.

اثر سمیت کادمیوم بر الگوی بیان ژن و فعالیت آنزیمی سوپراکسید دیسموتاز و آسکوربات پراکسیداز در گیاهچههای نخود (Cicer arietinum L.)
هنگامی که گیاهان در معرض شرایط محیطی تنشزا قرار میگیرند، تولید گونههای فعال اکسیژن (ROS) افزایش یافته و سبب خسارات عمده به سلولها میشود. در گیاهان سیستمهای دفاع آنتی اکسیدانی وجود دارند که میتوانند ترکیبات ROS را سمیتزدائی کنند. در این تحقیق تغییرات در الگوی بیان و فعالیت آنزیمی سوپراکسید دیسموتاز و آسکوربات پراکسیداز گیاهچههای نخود در پاسخ به غلظتهای مختلف کلرید کادمیوم (0، 5/0، 5/2، 5/7، 5/12، 25 و 50 میلی مولار) بررسی شد. نتایج نشان داد که در گیاه نخود تولید هر دو آنزیم SOD و APX در هر دو سطح mRNA و فعالیت آنزیمی در برگهای گیاهچهها در پاسخ به افزایش کادمیوم در محیط القاء میشوند. فعالیت آنزیمی SOD و APX و سطوح رونوشت ژنهای Cu/ZnSOD و APX در سطوح پائین و متوسط کادمیوم بطور معنیداری افزایش یافت ولیکن به دنبال افزایش غلظت کادمیوم از 5/12 به 50 میلی مولار فعالیت آنزیمی و میزان بیان دو ژن فوق به تدریج کاهش یافت. میتوان چنین نتیجهگیری کرد که سیستم دفاع آنتی اکسیدان آنزیمی در نخود نقش عمدهای در پاسخ به سمیت کادمیوم ایفا میکند.
