شناسائی میزبان های وحشی ویروس کوتولگی سبزرد هندوانه در جنوب و جنوب شرق ایران
ویروس کوتولگی سبزرد هندوانه (Watermelon chlorotic stunt virus, WmCSV) یکی از مخرب ترین ویروس های هندوانه در جنوب و جنوب شرقی ایران است. به منظور شناسائی علف هرزهای میزبان این ویروس، از مزارع بشدت آلوده هندوانه واقع در میناب (استان هرمزگان)، رودبار جنوب (جیرفت) و ارزوئیه (استان کرمان) بازدید گردید و نمونه برداری از داخل و حاشیه مزارع انجام شد. آلودگی و عدم آلودگی نمونه ها با آزمون PCR و با استفاده از آغازگرهای اختصاصی تعیین گردید. نتایج بدست آمده آلودگی تعداد زیادی از علف های هرز شامل چهارده گونه از دوازده جنس و نه تیره گیاهی را به WmCSV به اثبات رساند، بدون آنکه علائم خاصی روی بیشتر آنها دیده شود. تمامی این علف های هرز بعنوان میزبان WmCSV برای اولین بار معرفی می گردند. به منظور مقایسه جدایه های WmCSV در علف های هرز آلوده، قطعه 655 جفت بازی مربوط به ژن پروتئین پوششی 11 جدایه مختلف ویروس، همسانه سازی و تعیین ترادف گردید. مقایسه ترادف های نوکلئوتیدی و آمینواسیدی این جدایه ها با یکدیگر و با جدایه های موجود در بانک جهانی ژن نشان داد که جدایه های مورد مطالعه شباهت زیادی با یکدیگر و با سایر جدایه ها دارند. نزدیکترین جدایه WmCSV به جدایه های علف های هرز مورد مطالعه، جدایه بندرعباس (استان هرمزگان) بود که قبلا از این منطقه گزارش شده بود و از نظر ترادف های نوکلئوتیدی و آمینواسیدی بترتیب 2/99٪-9/98 و 100٪-8/96 با جدایه های فوق شباهت داشت. این نتایج نشان می دهد که این گیاهان بعنوان منابع نگاهدارنده WmCSV عمل کرده و بطور بالقوه می توانند در اپیدمیولوژی ویروس نقش اساسی داشته باشند.

مطالعه تنوع ژنتیکی تودههای بومی آفتابگردان آجیلی (Helianthus annuus L.) ایران با استفاده از نشانگرهای رتروترنسپوزونی IRAP
وجود تنوع ژنتیکی برای بدست آوردن ارقام با عملکرد بالا، کیفیت بهتر، تحمل بیشتر به تنشهای زیستی و غیرزیستی و مقاومت بیشتر به آفات و بیماریها، برای بهنژادگران ضروری است. رتروترنسپوزونها فراوانترین و رایجترین عناصر جابهجا شونده در ژنوم یوکاریوتها بهویژه ژنوم گیاهان میباشند. توزیع گسترده رتروترنسپوزونها در ژنوم گیاهان استفاده از آنها را به عنوان نشانگرهای مولکولی برای بررسی تنوع ژنتیکی ایدهآل ساخته است. در این مطالعه فعالیت رتروترنسپوزونها و همچنین تنوع ژنتیکی در 10 توده بومی آفتابگردان آجیلی با استفاده از نشانگرهای IRAP (Inter-retrotransposon amplified polymorphism) یررسی شد. از 25 آغازگر و ترکیب آغازگری IRAP، 11 آغازگر قادر به تولید الگوی باندی چند شکل با وضوح بالا بودند. 11 آغازگر IRAP، 116 مکان را تکثیر کردند که از این تعداد 110 مکان چند شکل بود. حداقل تشابه ژنتیکی نی (74/0) بین تودههای همدان و مشهد، و حداکثر تشابه (88/0) بین تودههای مرند و اصفهان مشاهده شد. تجزیه کلاستر با استفاده از آلگوریتم UPGMA، 10 توده بومی آفتابگردان آجیلی را در 3 گروه اصلی قرار داد. تجزیه واریانس مولکولی نشان داد تنوع درون تودهها بیشتر از تنوع بین تودها می باشد. بنابراین در برنامههای به نژادی آفتابگردان آجیلی، میتوان انتخاب را درون تودهها انجام داد. نتایج حاصل از واکنش زنجیره ای پلی مراز با نشانگرهای IRAP نشان داد که رتروترنسپوزونهای مطالعه شده در ژنوم آفتابگردان فعال هستند و به صورت سربهسر، سربهدم و دمبهدم در ژنوم ادغام شدهاند. نتایج حاصل از مطالعه حاضر نشان می دهد که نشانگرهای مبتنی بر رتروترنسپوزون ها ابزاری مفید برای مطالعات ژنومیکس در آفتابگردان میباشند.

بررسی اثر محلول پاشی بنزیل آدنین قبل از شکوفایی گل در توسعه تخمک و نجات جنین دو رقم انگور بکربار کاذب ایرانی (Vitis vinifera L.)
یکی از مهمترین اهداف اصلاحی انگورهای رومیزی تولید دورگهای بیدانه جدید با حبههای درشت، سفت، معطر و مناسب با سلیقه مصرف کنندگان میباشد. سقط جنین در ارقام بیدانه به طور عمده کارآیی اصلاح ارقام بیدانه را محدود میکند. بر اساس اینکه سیتوکینینها موجب افزایش قدرت مقصد فیزیولوژیکی دانه ها برای اسیمیلاتها میشوند، تحقیق حاضر به منظور مطالعه اثر محلولپاشی بنزیل آدنین قبل از شکوفایی گل در توسعه تخمک و نجات جنین دو رقم انگور بکربار کاذب( عسکری و بیدانه سفید) انجام گرفت. غلظتهای بنزیل آدنین صفر،60 و100 میلی گرم در لیتر بود. تخمکهای رقم بیدانه سفید،20 روز و رقم عسکری 40 روز بعد از باز شدن گل از درون حبهها بیرون آورده شد و سپس بر روی محیط کشت نیچ و نیچ حاوی 1 میکرومول جیبرلین،1 میکرومول نفتالین استیک اسید، 2 گرم در لیتر زغال فعال ،20 گرم در لیتر ساکارز و 8 گرم در لیترآگار کشت شدند. صفات مورد بررسی شامل تخمکهای قهوهای شده، کالوس داده و جوانهزده بود. محلولپاشی بنزیل آدنین اثر معنیدار بر قهوهای شدن تخمکها نداشت اما درصد تخمکهای قهوهای شده در بین ارقام مورد آزمایش معنیدار بود و بیشترین تخمک قهوهای شده در رقم بیدانه سفید مشاهده گردید. اثر محلولپاشی بنزیل آدنین با غلظتهای 60 و100 میلیگرم در لیتر بر درصد کالوسدهی و جوانهزنی تخمکها معنیدار بود. بر اساس نتایج بدست آمده درصد تخمکهای جوانه زده، بین دو رقم مورد آزمایش معنیدار بود. تخمکهای جوانهزده در رقم عسکری71/23 درصد و در رقم بیدانه سفید44/14 درصد بود. بطور کلی محلولپاشی با بنزیل آدنین قبل از گلدهی باعث افزایش موفقیت تکنیک نجات جنین در ارقام عسکری و بیدانه سفید میشود

بررسی ارتباط بین چند شکلیهای موجود در چهار ژن کاندیدا با صفات تولید شیر و عملکرد تولید مثلی گاوهای سرابی با استفاده از نشانگر PCR-RFLP
انتخاب برای بهبود صفات تولید و تولید مثل در دامها، امری زمانبر بوده و فقط در سنین بلوغ دامها امکانپذیر است. گاو سرابی یکی از نژادهای بومی شیری در ایران است که به طور عمده در شمالغرب کشور پرورش مییابد. شناسایی ژنهای کاندیدای مؤثر بر صفات تولید شیر و عملکرد تولید مثلی در این نژاد دارای اهمیت بوده و میتواند بازده برنامههای اصلاح نژادی را بهبود دهد. هدف از مطالعه حاضر بررسی ارتباط بین چندشکلی موجود در چهار ژن کاندیدا (لپتین، هورمون رشد (GH)، گیرندهی هورمون رشد (GHR) و ژن pit-1) با صفات تولید شیر و برخی صفات تولید مثلی گاو سرابی با استفاده از نشانگر pcr-rflp بود. نتایج نشان داد که بیشترین فراوانیهای آللی برای هر یک از ژنهای لپتین، GH، GHR و pit-1 به ترتیب برای آللهای A (72/0)، V(51/0)، + (65/0) و B (55/0) بود. ژنوتیپهایll ، -/-، ab-bb مربوط به ژنهای GH، GHR و pit-1 نسبت به سایر ژنوتیپها به طور معنیداری تولید شیر بیشتری داشتند (p< 0.01). همچنین ژنوتیپهای ll و AB شناسایی شده در ژنهای GH و pit-1 نسبت به سایر ژنوتیپها روزهای باز کمتری را نشان دادند (p< 0.01). در مطالعه حاضر هیچ رابطه معنیداری بین ژنوتیپ این ژنها و سایر صفات بررسی شده مشاهده نشد. در ژنهای کاندید مذکور مقادیر هتروزیگوسیتی مشاهده شده نسبت به مقادیر مورد انتظار کمتر بودند و نتایج تجزیه و تحلیل کایمربع نشان دهندهی عدم وجود تعادل هاردی- واینبرگ در این جایگاهها بود. به طور کلی نتایج این تحقیق نشاندهنده وجود ارتباط بین جهشهای موجود شناسائی شده در برخی از این ژنها با صفات تولید شیر و روزهای باز در گاوهای سرابی است و امکان استفاده از این اطلاعات در برنامهی انتخاب به کمک نشانگرها باید مورد بررسی بیشتر قرار گیرد.

تعیین توالی ژنوم کامل و فیلوژنی جدایه های نژاد شدید ویروس پیچیدگی زرد برگ گوجه فرنگی جدا شده از تاتوره Datura stramomum L. در منطقه بجنورد، ایران
ویروس پیچیدگی زرد برگ گوجه فرنگی , TYLCV) Tomato yellow leaf curl virus) به همراه ده گونه ویروسی و نژادهایشان که به ویروسهای شبه TYLCV معروفند عامل خسارت به گوجه فرنگی (Solanum lycopersicum L.) در مناطق گرمسیر، نیمه گرمسیر و معتدل دنیا می باشند. در این بررسی تعداد سه جدایه TYLCV-IL از گیاهان تاتوره در منطقه بجنورد همسانه سازی و ژنوم کامل آنها تعیین توالی گردید. توالی اسید نوکلئیک ژنوم کامل آنها به همراه تعداد نه جدایه TYLCV-IL گزارش شده ازایران و هشت توالی سایر ویروسهای Begomovirus گوجه فرنگی از ایران و سایر مناطق دنیا مورد تجزیه و تحلیل قرار گرفتند. نتایج فیلوژنی جدایهها نشان داد که تکامل جمعیت نژاد TYLCV-IL در ایران وابسته به جغرافیا بوده و صرفنظر از گونه میزبان واریانتهای شمال شرق و جنوب ایران به طور مجزا در دو زیرگروه جداگانه قرار گرفتند. جدایه TYLCV-IL[IR:Sh40:07], GUO76444 از منطقه شیراز با هیچکدام از جمعیتهای شمال شرق و جنوب ایران گروه بندی نشد. این جدایه با نژاد تیپ TYLCV-IL گزارش شده از فلسطین اشغالی قرابت نزدیکی داشته و با یکدیگر در یک زیر گروه مجزا قرار گرفتند. نتایج تجزیه فیلوژنی نشان داد که این جدایه از نظر تکاملی به عنوان پل ژنتیکی بین جدایههای TYLCV-IL حوزه مدیترانهای و ایران محسوب میشود. این اولین گزارش از گیاه تاتوره به عنوان میزبان نژاد TYLCV-IL در ایران و همچنین حضور این نژاد ویروسی در منطقه بجنورد و استان خراسان شمالی میباشد

فسفر یکی از عناصر غذایی مهم برای گیاهان میباشد که در خاک فراهمی کمی دارد. فسفر در خاک به دو شکل آلی و معدنی یافت میشود. باکتریهای تحریککننده رشد گیاه یا PGPR، باکتریهای موجود در خاک و ریزوسفر گیاهان هستند که با سازوکارهای مختلف به رشد گیاه کمک میکنند. توانایی برخی ریزسازوارهها به منظور تبدیل فسفر نامحلول به شکل قابل استفاده مانند ارتوفسفات، ویژگی مهمی در PGPR میباشد که باعث افزایش عملکرد گیاهان میشود. گونههایی از جنس Pseudomonas، Bacillus، Pantoea وRhizobium از قویترین حلکنندگان فسفات به شمار میآیند. سازوکار اصلی برای انحلال فسفات معدنی تولید اسیدهای آلی است و در انحلال اشکال فسفر آلی اسید فسفاتازها نقش اصلی را در خاک بازی می کنند. روش مرسوم در جداسازی این دسته از باکتریها استفاده از منابع فسفاته معدنی و آلی کم محلول یا نامحلول در محیط کشتهای مایع یا جامد میباشد، که از طریق پایش تولید فسفات آزاد شده و کاهش pH در محیط مایع یا مشاهده هاله شفاف در اطراف کلنیها و تولید کلنیهای رنگی (سبز ، آبی و زرد) در صورت استفاده از سوبستراهای رنگزا در محیط کشت جامد دنبال میشود. گر چه دانش ژنتیک انحلال فسفات هنوز اندک میباشد، چندین ژن رمزکنندة فسفاتاز مشخص گردیده و همسانهسازی شدهاند و تعدادی ژن درگیر در انحلال فسفات معدنی جداسازی شده است. روشهای زیستشناسی مولکولی رویکردی مفید برای به دست آوردن و تشخیص سویههای PGPR کاآمد میباشد. انتقال و بیان ژنهای درگیر در انحلال فسفات (فسفات آلی یا معدنی) در باکتریها یا گیاهان یک راهکار جدید برای بهبود ظرفیت ریزسازوارهها به عنوان مایه تلقیح میکروبی است.

شناسایی جایگاه های ژنی مؤثر بر نسبت کلیبر در یک جمعیت از آمیخته های حاصل از تلاقی دو سویه بلدرچین ژاپنی
به منظور شناسایی QTL مؤثر بر صفات رشد و برآورد میزان اثر آنها، از یک طرح تلاقی F2 حاصل از دو سویه متفاوت بلدرچین ژاپنی (تخمگذار یا سفید و گوشتی یا وحشی) استفاده گردید. با تلاقی 34 پرنده حاصل از تلاقی فوق و به منظور ایجاد جمعیت لازم برای رکورد برداری فنوتیپی، 422 پرنده نسل سوم تولید شد. مشاهدات شامل وزن های هفتگی ثبت شده و نسبت کلیبر مربوط به فاصله زمانی هچ تا 1، 1 تا 2، 2 تا 3، 3 تا 4، 4 تا 5 و هچ تا 5 هفتگی می باشد. تمامی پرندگان هر سه نسل (472 پرنده) برای 8 نشانگر ریزماهواره موجود بر روی کروموزوم شماره 1 و با میانگین فاصله 29 سانتی مورگان از یکدیگر تعیین ژنوتیپ شدند. آنالیز داده های حاصل با استفاده از روش نقشه یابی درون فاصله ای مبتنی بر رگرسیون انجام گرفته و پنج QTL مربوط به نسبت کلیبر محاسبه شده برای 1 تا 2، 3 تا 4، 4 تا 5 و هچ تا 5 هفتگی شناسایی شد. واریانس QTLهای شناسایی شده برای صفات مختلف در محدوده 8/0 تا7/3 بود. با برازش مدل دارای اثرات افزایشی و غلبه، اثر غلبه برای هیچکدام از صفات معنی دار نشد. همچنین، جایگاه های شناسایی شده، بر هیچکدام از صفات مورد بررسی اثر ایمپرینتینگ نداشتند.

بررسی گیاهان ترانسپلاستومیک نسل اول (T1)توتون حاوی ژن اینترفرون گامای انسانی (hIFNg)
در دهههای اخیر، پیشرفت در بیوتکنولوژی گیاه، بویژه زراعت مولکولی امکان استفاده از گیاهان را به عنوان یک سیستم تولید جدید و راکتور زیستی برای محدودهی وسیعی از پروتئینهای نوترکیب فراهم کرده است. اینترفرون گامای انسانی یکی از پروتئینهای ارزشمند دارویی میباشد که کاربردهای پزشکی وسیعی در زمینههای تشخیصی و درمانی پیدا کرده است. کلونینگ ژن اینترفرون گامای انسانی به همراه ژن مقاومت به آنتیبیوتیک اسپکتینومایسین - استرپتومایسین با استفاده از ناقل کلروپلاستی pKCZ در آزمایشگاه بیوتکنولوژی دانشکده کشاورزی دانشگاه تربیت مدرس صورت گرفته و سپس به روش بیولستیک به کلروپلاست گیاه توتون انتقال یافته است. هدف از انجام این تحقیق بررسی پایداری بیان ژن اینترفرون گامای انسانی در گیاهان ترانسپلاستومیک نسل اول توتون بود. گیاهان رشد کرده در سطح محیط انتخابگر، در سطوح مختلف (DNA، RNA و پروتئین) به منظور تایید درج و بیان ژن مورد بررسی قرار گرفتند. بررسی این گیاهان در سطح DNA با استفاده از آغازگرهای اختصاصی ژن اینترفرون گاما و تکنیک PCR صورت پذیرفت، نتایج حاصل درج این ژن را در ژنوم کلروپلاستی گیاهان تایید کرد، به علاوه با آزمون RT-PCR رونویسی ژن هدف تایید شد. همچنین به منظور بررسی بیان ژن هدف در سطح پروتئین، آزمونهای SDS-PAGE، Dot-blot و Western-blot انجام شد و نتایج بدست آمده، بیان این ژن را در سطح پروتئین تایید کرد. در نهایت به منظور تعیین میزان بیان ژن اینترفرون گامای انسانی آزمون ELIZA انجام شد. نتایج نشان داد که بالاترین سطح تجمع اینترفرون گاما در گیاهان ترانسپلاستومیک بالای 2/0% پروتئین محلول کل میباشد.

مطالعه اثر ژن Rf3 برای بازگردانندگی باروری به برنج در زمینه ژنتیکی لاین ندا-A
در برنامه تولید بذر هیبرید در سیستم سه لاینی، استفاده از لاین با قابلیت ترکیبپذیری خصوصی مطلوب حامل ژنهای بازگرداننده باروری (Rf) ضرورت دارد. در این تحقیق، جایگاه ژنی بازگرداننده باروری Rf3 واقع بر روی کروموزوم شماره یک برنج با روش تلاقی برگشتی به کمک نشانگر، به پسزمینه ژنتیکی لاین نرعقیم ندا-A انتقال داده شد و همزمان اثر آن در هر نسل بر باروری دانه گرده و خوشه لاین گیرنده، مورد بررسی قرار گرفت. جهت انتقال، یک گیاه از جمعیت نسل F2 (حاصل از تلاقی میان ندا-A و IR36) بر اساس نشانگرهای پیوسته به جایگاه ژنی Rf3 (RM1، RM3873 و RM3233) انتخاب و با لاین ندا-A (به عنوان والد تکراری) تلاقی برگشتی داده شد. نتاج نسل اول تلاقی برگشتی (BC1) از نظر وجود نشانگرهای پیوسته به ژن و همچنین از نظر فنوتیپی و به دنبال آن برای 15 نشانگر ریز ماهواره پسزمینه، مورد بررسی قرار گرفتند. تنها دو بوته نسل BC1 که در هر سه جایگاه نشانگری دارای آلل غالب از والد بخشنده بودند باروری بالای خوشه (55% و 65%) را نشان دادند. نتاج نسل دوم تلاقی برگشتی (BC2) از تلاقی دو بوته فوق با والد تکراری حاصل شد. در بین نتاج BC2 یک گیاه با باروری بالاتر خودگشن شده و 170 بوته از نسل BC2F2 از نظر 3 نشانگر RM1، RM3873 و RM3233 و همچنین دانهبندی خوشه مورد بررسی قرار گرفتند. هفت بوته با میزان بالاتری از ژنوم والد تکراری (از 1/91 % تا 5/98%) و در وضعیت هموزیگوتی کامل از نظر سه نشانگر پیوسته به Rf3 (RM1، RM3873 و RM3233) شناسایی شدند که دارای باروری بالای دانه گرده و همچنین خوشه (75%تا 97%) بودند که نشان میدهد هر چه هموزیگوسیتی جایگاه ژنی Rf3 بیشتر شد، بازگرداندن باروری به نرعقیمی نوع وحشی (WA) در پسزمینه ژنتیکی لاین گیرنده نرعقیم، بیشتر تقویت گردید. بنابراین، میتوان نتیجهگیری نمود که جایگاه ژنی Rf3 تاثیر متقابلی با نرعقیمی سیتوپلاسمی نوع WA در جهت افزایش باروری دانه گرده و خوشه برنج دارد و از اینرو، میتواند همراه با سایر ژنهای بازگرداننده باروری در برنامه تولید بذر هیبرید برنج به کار گرفته شود.

شناسایی تنوع ژنتیکی و بررسی توالی نوکلئوتیدی ناحیه پروموتر ژن گیرنده هورمون رشد در گاوهای بومی استان کهکیلویه و بویر احمد
پروتئین گیرنده هورمون رشد یک گیرنده سطح سلولی برای هورمون رشد است که جهت انجام نقش هورمون رشد در بافت های هدف ضروری است. هدف تحقیق حاضر، شناسایی و بررسی تنوع ژنتیکی ناحیه پروموتر ژن گیرنده هورمون رشد درگاوهای بومی استان کهکیلویه و بویراحمد می باشد. برای این منظور، ناحیهای از پروموتر ژن گیرنده هورمون رشد توسط تکنیک تفاوت فرم فضایی رشته های منفرد (SSCP) و تعیین توالی در گاوهای نژاد بومی تعیین ژنوتیپ شد. در کل 50 نمونه بررسی شده 3 الگوی (SSCPs) مجزا مشاهده گردید که با تعیین توالی نوکلوتیدی آنها چند شکلی تک نوکلئوتیدی A/G در جایگاه 154- و دو طول متفاوت 17 و 11 تکراری برای ریزماهواره TG در جایگاه 86- قبل از شروع رونویسی مشاهده گردید. فراوانی ژنوتیپهای AA (17/17TG)، GG (11/11TG) و GA (11/17TG) در جایگاه 154- به ترتیب 34/0، 24/0 و 52/0 بود. بررسی بیوانفورماتیکی چند شکلیهای شناسایی شده بر روی جایگاه فاکتورهای رونویسی نشان داد که چند شکلی تک نوکلئوتیدی در جایگاه 154- بسیار نزدیک به جایگاه اتصال برای فاکتور رونویسی C/EBPb قرار دارد که ممکن است فرآیند رونویسی ژن را تحت تاثیر قرار دهد. نقش کاربردی ممکن چندشکلی های شناسایی شده باید توسط تجزیه و تحلیل بیان ژن تایید گردد.

تولید فرم نوترکیب ایزوفرم تیپ 1 متالوتیونین برنج (OsMTI-1b) در باکتری اشریشیاکلی و بررسی قابلیت اتصال آن به فلز نیکل
گیاهان به مکانیسمهای سلولی مختلفی در مقابله با اثر سمی فلزات سنگین مجهز شدهاند. یکی از مهمترین این مکانیسمها، تولید پروتئینها و پپتیدهای متصل شونده به فلزات، مانند متالوتیونینها (MT) میباشد. متالوتیونینها، گروهی از پروتئینها با وزن مولکولی کم و غنی از آمینواسیدهای سیستئین هستند که دارای ظرفیت بالای اتصال به فلزات میباشند. در این تحقیق توالی کد کنندهی ایزوفرمOsMTI-1b از گیاه برنج، به عنوان گیاه مدل در بین تک لپهایها، در ناقل بیانی pET41a همسانهسازی و به میزبان بیانی E. coli سویهی Rosetta (DE3) منتقل شد. پس از القا محیط کشت توسط IPTG، میزان مناسبی از پروتئینهای نوترکیب در فاز محلول تولید و با استفاده از روش کروماتوگرافی جذبی خالص سازی شد. سلولهای باکتری بیانکنندهی پروتئینهای نوترکیب در محیط LB حاوی نمک NiCl2 رشد داده شدند و منحنی رشد آنها در مقایسه با شاهد ترسیم شد. مقدار کاهش فلز نیکل در محیط کشت و تجمع آنها در رسوب باکتریایی توسط دستگاه طیف سنجی پلاسمای جفت شدهی القائی (ICP-AES) بهدست آمد. بر اساس نتایج مشخص گردید بیان ایزوفرمOsMTI-1b از طریق افزایش تجمع درون سلولی فلز نیکل موجب افزایش تحمل باکتری E. coli به این فلز میگردد. همچنین، بررسی الگوی جذب نور و واکنش DTNB با پروتئینهای انکوبه شده با فلز نیکل در شرایط این ویترو، تشکیل دستجات فلز/تیول در پروتئین نوترکیب GST-OsMTI-1b را اثبات نمود. یافتههای پژوهش حاضر میتواند منعکس کننده نقش احتمالی ایزوفرمOsMTI-1b در تحمل گیاه برنج به تنش فلزات سنگین باشد.

اثرات پارامترهای مختلف بر روی کیفیت نتایج حاصل از تکنیک cDNA-AFLP در مطالعه ترانسکریپتوم سیب زمینی
آنالیزهای ترانسکریپتوم در ایجاد اطلاعات و درک کافی از سیستم های واکنشی گیاهان در برابر عوامل زیستی و غیر زیستی بسیار کارآمد هستند. cDNA-AFLP به عنوان روشی مناسب علاوه بر تکرار پذیری بالا، نیاز به اطلاعات اولیه در مورد توالی ژن ها ندارد. البته عوامل متعددی در حساسیت، دقت و تکرار پذیری آن موثر هستند که توجه به این عوامل می تواند باعث افزایش کیفیت ارزیابیها و دستیابی به اطلاعات صحیح گردد. در تحقیق حاضر cDNA-AFLP برای آنالیز واکنش گیاه سیب زمینی در برابر استرس ناشی از باکتری بیماریزای Ralstonia solanacearum در شرایط in vitro بهینه سازی گردید. در این راستا پارامترهای مختلف و اثر تعامل آنها با یکدیگر در افزایش حساسیت و تکرار پذیری این روش بررسی شد. نتایج نشان داد روش استخراج RNA و استفاده از mRNA از فاکتور های تاثیر گذار در موفقیت آزمون cDNA-AFLP است به طوری که RNA های استخراج شده با استفاده از روش مبتنی بر ستون از یکنواختی و کیفیت بالاتری برخوردار هستند و استخراج mRNA علاوه بر حذف RNA های ریبوزومی، در کاهش ممانعت کننده ها و اجرای صحیح سایر مراحل آزمون تاثیر گذار است. همچنین بهینه سازی فاکتورهایی از قبیل رقت مناسب محصول پیش تکثیر که به عنوان الگو در مرحله تکثیر انتخابی استفاده میشود، تعداد سیکل PCR در مرحله پیش تکثیر، غلظت مناسب MgCl2 و Taq DNA Polymerase در مراحل پیش تکثیر و تکثیر انتخابی، نقش تعیین کننده ای بر روی تعداد و غلظت قطعات cDNA تکثیر شده و وضوح آن بر روی ژل دارد.

بررسی تنوع ساختار کروموزومی در ژنوم گاوهای هلشتاین با استفاده از بسته نشانگری 50K
تنوع ساختار کروموزومی در مکانیزمهای بیولوژیکی از اهمیت خاصی برخوردار است. به منظور تعیین تعداد و توزیع این تنوع در ژنوم سه کروموزم 6، 14 و 20 که حضور جایگاه صفات کمی موثر بر تولید در آن تایید شده است٬ مورد بررسی قرار گرفت. به این منظور از 580 راس گاو هلشتاین نمونه خون گرفته شد و پس از استخراج DNA٬ تعیین ژنوتیپ با استفاده از بسته نشانگری 50K گاوی شرکت ایلومینا صورت گرفت. پس از تصحیح دادهها برای شدت سیگنال و نواحی غنی از GC، تعداد 383 نمونه برای آنالیز نهایی باقی ماند. دادهها بر اساس اسمبلی UMD3.1 ژنوم گاو برای کروموزمهای 6، 14 و 20 آنالیز شد. پس از اعمال کلیه فیلترها تعداد 199 تنوع ساختار کروموزومی (132 حذف و 67 اضافه) با متوسط 5/0 برای هر فرد و میانگین طول kb 3/147 و میانه kb 4/139 شناسایی شد. نسبت حذف به اضافه برابر با 97/1 و کروموزمهای 6 و 20 به ترتیب دارای بیشترین و کمترین تنوع ساختار کروموزومی بودند. آنالیزهای بیوانفورماتیکی مشخص نمود که 77 ژن با این نواحی تنوع ساختار کروموزومی در ارتباط هستند. نتایج این پژوهش نشان داد تنوع ساختار کروموزومی بخش قابل توجهی از این سه کروموزم که حاوی تعداد قابل توجهی جایگاه صفات کمی هستند را پوشش میدهد. به دلیل اثر این تنوع بر ایجاد تغییر در ساختار و دز ژنها، به نظر میرسد تنوع ساختار کروموزومی میتواند بر واریانس فنوتیپی صفات کمی موثر باشد لذا احتمالا این تنوع قابلیت استفاده در برنامههای اصلاحنژادی را دارد.

بررسی تنوع ژنتیکی ارقام گندم نان (Triticum aestivum L.) از طریق صفات مورفوفیزیولوژیک و نشانگرهای مولکولی SSR
وجود تنوع ژنتیکی شرط ضروری برای موفقیت در برنامههای بهنژادی است. به منظور بررسی تنوع ژنتیکی از طریق برخی از صفات مورفوفیزیولوژیکی و نشانگرهای SSR، 40 رقم گندم نان در یک طرح بلوک کامل تصادفی با 3 تکرار در مزرعه پژوهشی دانشگاه شهید باهنر کرمان در سال 1390-1389 مطالعه شدند. ژنوتیپهای گندم نان برای تمامی صفات مورد بررسی تنوع ژنتیکی قابل ملاحظهای نشان دادند. تعداد پنجههای بارور بیشترین همبستگی را با عملکرد دانه نشان داد. در آزمایش SSR از مجموع 10 آغازگر انتخابی، 9 آغازگر چند شکلی قابل توجهی نشان دادند. برای مجموع ژنوتیپ ها 31 نوار چند شکل با میانگین 4/3 نوار به ازای هر آغازگر مشاهده شد. هتروزیگوسیتی مورد انتظار کل جمعیت در تمام جایگاه های ژنی به طور متوسط 36/0 ارزیابی شد و جایگاه ژنی wmc 420 دارای بیشترین تنوع بر روی جمعیت مورد مطالعه بود. تجزیه کلاستر با استفاده از روش وارد انجام شد و ارقام بر اساس میزان شباهتها گروهبندی شدند. اطلاعات این گروه بندی میتواند در پروژههای بهنژادی برای افزایش عملکرد در شرایط تنش مورد استفاده قرارگیرد.
