بررسی میزان هورمونهای گل سرخ تراریخته با ژن etr1-1 تحت تیمارهای اتیلن و جیبرلین
هدف: گل سرخ از مهمترین گلهای شاخه بریده دنیا بوده که تا کنون مطالعات زیادی برای حفظ کیفیت و ماندگاری پس از برداشت آن انجام شده است. از عوامل اصلی محدود کننده ماندگاری در مرحله پس از برداشت میتوان به هورمون اتیلن اشاره نمود. لذا دستورزی ژنتیکی به منظور کاهش اثرات نامطلوب این هورمون مورد توجه بوده و در پژوهش حاضر، گلهای سرخ تراریخته حاوی ژن جهشیافته etr1-1 مورد ارزیابی قرار گرفتند. همچنین با توجه به اثرات آنتاگونیستی، تغییر غلظت هورمون جیبرلین نسبت به اتیلن به نفع بالاتر بردن غلظت جیبرلین در مطالعه اخیر مورد توجه قرار گرفت. مواد و روشها: میزان فیتوهورمونهای لاین ماندگار (تراریخته) و شاهد (غیر تراریخته) پس از اعمال تیمارهای اتیلن (صفر وL L-1µ 1) و جیبرلین (mg L-1 ۸۰) در مراحل غنچه تجاری و نیمه باز بررسی شدند. اعمال تیمار اتیلن با تزریق گاز خالص به داخل کیسههای پلیاتیلنی توسط سرنگ انجام شد. جهت اعمال تیمار جیبرلین، گلهای شاخه بریده در محلول جیبرلین با غلظت مورد نظر قرار داده شدند. تیمارها به مدت ۲۴ ساعت اعمال گردید و نمونهبرداری از بیرونیترین ردیف گلبرگها انجام شد. آزمایشها بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با چهار تکرار به منظور اندازهگیری میزان فیتوهورمونها اجرا شد. نتایج: نتایج نشان داد که کاهش در میزان هورمونهای جیبرلین، بنزیل آدنین و ایندول استیک اسید و افزایش در میزان هورمون آبسیزیک اسید و اتیلن در مراحل غنچه و نیمهباز در لاین ماندگار نسبت به شاهد معنیدار بود. همچنین، بیشترین میزان هورمونهای جیبرلین، بنزیل آدنین و ایندول استیک اسید و کمترین میزان اتیلن و آبسیزیک اسید مربوط به لاین ماندگار در تیمار با جیبرلین بود. نتیجهگیری: با توجه به نتایج موجود به نظر میرسد که ژن etr1-1 میتواند کاندیدای مناسبی، جهت به تأخیر انداختن فرایند پیری وابسته به اتیلن در گلهای حساس باشد که همراه با تیمار جیبرلین از طریق کاهش خسارت اکسیداتیو ماندگاری را بهطور قابل توجهی افزایش میدهد.

ردیابی ژن های مقاوم به بیماری آتشک درختان سیب (Malus domestica) استان اصفهان با استفاده از نشانگرهای SCAR و SSR
هدف: بیماریهای گیاهی میتوانند عامل محدود کننده کاشت یک گیاه در یک منطقه باشند. یکی از شیوههای جدید در تولید گیاهان مقاوم به بیماریها، استفاده از نشانگرهای مولکولی میباشد. نشانگرهای مولکولی قادر به کشف و آنالیز ژنهای مهم مقاومت هستند. لذا با توجه به شدت خسارت و تنوع ژنتیکی بیماری آتشک که از بیماریهای خطرناک درختان میوهی دانهدار است، ارزیـابی ژرمپلاسـم درخت سیب ضروری میباشد. از این رو پژوهشی با هدف ردیابی ژنهای مقاومت به بیماری با استفاده از 6 نشانگر SCAR و SSR در برخی ژنوتیپهای سیب استان اصفهان انجام شد. مواد و روشها: از برگهای جوان70 نمونه سیب استان اصفهان، در اوایل اردیبهشتماه نمونهبرداری و DNA به روش CTAB استخراج گردید. واکنش زنجیره ای پلیمراز جهت تکثیر قطعات ژنومی6 جفت آغازگر انجام شد. نتایج: نتایج نشان داد در جمعیتها، آغازگرها باند نادر و معمولی تکثیر شده در 25 یا کمتر از 25 درصد جمعیت و 50 یا کمتر از 50 درصد جمعیت تکثیر نکردند. جمعیت سمیرم- حنا بیشترین مقدار شاخصهای تنوع ژنتیکی نی، شانون، تعداد آلل موثر و متفاوت را داشت که نشان دهنده تنوع ژنتیکی بیشتر در این جمعیت نسبت به سایر جمعیتها است. تجزیه واریانس مولکولی نشان داد تنوع بین جمعیتی از لحاظ آماری معنیدار نمیباشد و 92% تنوع مربوط به درون جمعیتها بود. بیشترین فاصله ژنتیکی بین جمعیتهای سمیرم-حنا و سمیرم- پادنا بود و فاصله بین دورترین و نزدیکترین جمعیتها کم بود که این نتیجه به وسیله تجزیه واریانس مولکولی نیز تایید گشت. نتیجهگیری: 6 آغازگر به خوبی تکثیر شده و تمایز بین جمعیت ها را نشان دادند لذا نشانگرها به درستی انتخاب شده اند. همچنین نتایج حاکی از تنوع بین جمعیتی بسیار کم بود اما با تلاقی دورترین و نزدیکترین جمعیت و سپس تلاقی نتاج بدست آمده، میتوان جهت هرمی شدن ژنهای مورد نظر و تولید نوترکیبی قویتر و متنوعتر بهره جست.

ارتباط نشانگری و تنوع ژنتیکی صفات زراعی گلرنگ (Carthamus tinctorius L.) با استفاده از نشانگر AFLP
هدف: این آزمایش با هدف بررسی تنوع ژنتیکی، تعیین بهترین ساختار ژنتیکی و تجزیه ارتباطی گلرنگ با استفاده از نشانگر AFLP جهت شناسایی نشانگرهای پیوسته با صفات زراعی مختلف انجام شد.
مواد و روشها: در این مطالعه 17 ژنوتیپ گلرنگ به صورت طرح بلوک کامل تصادفی با 3 تکرار در مزرعه تحقیقاتی دانشگاه شهید باهنر کرمان در سال 1395 کشت گردیدند. صفات فنوتیپی شامل عملکرد دانه، ارتفاع بوته، تعداد قوزه در بوته، تعداد دانه در قوزه، وزن هزار دانه، قطر قوزه، روز تا 50% گلدهی و روز تا رسیدگی اندازهگیری شد. تکنیک AFLPبا استفاده از هشت ترکیب آغازگری EcoRI و MseI انجام شد.
نتایج: در کل 147 باند چند شکل با میانگین 58/81 درصد چندشکلی ایجاد شد. تجزیه کلاستر بر اساس روش الگوبندی UPGMA و معیار جاکارد ژنوتیپهای گلرنگ را به دو گروه تقسیم کرد. تعداد 45 و 39 نشانگر به ترتیب بر اساس مدل GLM و MLM ارتباط معنیدار با صفات مورد مطالعه داشتند. نشانگرهای M14/E6-10، M14/E11-16، M14/E11-13، M3/E10-14 و M4/E36-12 با عملکرد دانه، M3/E10-12، M3/E36-29، M59/E36-21 و M14/E11-10 با تعداد قوزه در بوته، M4/E36-8، M59/E36-21، M3/E10-9، M14/E11-14 و M14/E11-13 با تعداد دانه در قوزه، M4/E36-19،12 M4/E36-، M14/E11-1 و M4/E10-1 با وزن هزار دانه، M4/E10-2، M59/E36-21، M3/E36-30 و M4/E36-24 با ارتفاع گیاه، M3/E36-24، M3/E36-6، M3/E10-20 و M4/E36-18 با قطر قوزه، M14/E11-10، M3/E36-30 و M59/E36-21 با روز تا 50% گلدهی و M14/E11-10 و M59/E36-21 با روز تا رسیدگی در هر دو مدل همبستگی معنیدار نشان دادند.
نتیجهگیری: نشانگرهای AFLP مشخص شده با اثرات قوی در این مطالعه میتواند کاندیدهای مناسبی برای انتخاب به کمک نشانگر در برنامههای اصلاحی و تبدیل به نشانگرهای اختصاصی دیگر باشند.

تاثیر محرکهای ساکارز و کیتوزان بر بیان ژن کد کننده آنزیم CYP79 F1 و محتوی سولفورافان در گیاهچههای ازمک
هدف: گلوکورافانین، یک گلوکوزینولات الیفاتیک است که در گیاه ازمک (Lepidium draba)از خانواده شببو به فراوانی یافت میشود. این گلوکوزینولات در حضور آنزیم میروزیناز به ایزوتیوسیانات سولفورافان تبدیل میشود که فعالیتهای زیستی مختلفی از قبیل خاصیت آنتیاکسیدانی و آنتی باکتریایی و همچنین توانایی مهار رشد و تکثیر سلولهای سرطانی را دارد. CYP79 F1 اولین آنزیم در مسیر بیوسنتز گلوکورافانین است.
مواد و روشها: در این مطالعه، گیاهچههای ازمک با سه تکرار مستقل و در قالب طرح کاملاً تصادفی بمدت 7 روز در حضور غلظتهای مختلف ساکارز و کیتوزان (صفر، 25، 50، 100، 200 و 400 میلیگرم بر لیتر) رشد کردند. پس از جمع آوری گیاهچهها، محتوای سولفورافان در اندام هوایی گیاهچهها با استفاده از دستگاه HPLC اندازهگیری شد. علاوه بر این، میزان بیان ژن CYP79 F1 در گیاهچههای تیمار شده با استفاده از تکنیک Real Time PCR مورد آنالیز قرار گرفت.
نتایج: نتایج نشان داد که محتوی سولفورافان در گیاهچههای تیمار شده با ساکارز با افزایش غلظت آن در محیط بهطور معنیداری نسبت به نمونه شاهد افزایشیافته است، درحالیکه در گیاهچههای تیمار شده با کیتوزان، افزایش معنیدار محتوی سولفورافان فقط در غلظت mg/L200 مشاهده گردید. نتایج حاصله نشان داد که بیان ژن CYP79 F1 در گیاهچههای تیمارشده با ساکارز در غلظت 50 میلیگرم بر لیتر و با کیتوزان در غلظتهای 50 و 100 میلیگرم بر لیتر نسبت به نمونه شاهد بهطور معنیداری افزایشیافته است.
نتیجهگیری: براساس نتایج بدست آمده در این تحقیق، چنین نتیجهگیری میشود که غلظتهای مذکور این محرکها میتوانند با اثر بر بیان ژن CYP79F1 بیوسنتز گلوکوزینولاتهای آلیفاتیک را تحریک نمایند. با توجه به نقش و اهمیت سولفورافان به عنوان یک متابولیت دارویی باارزش و همچنین نقش آنزیم CYP79F1 در سنتز گلوکورافانین، پیشنهاد میشود تاثیر الیسیتورهای مذکور بر تولید سولفورافان و بیان ژن CYP79F1 در غلظتها و زمانهای دیگر نیز بر روی این گیاه مورد آنالیز قرار گیرد.

بهینهسازی کشت بافت و انتقال سازه حاوی ژن GME کیوی به گیاه کاهو (Lactuca sativa L. )
هدف: کاهو به دلیل سازگاری با کشت بافت و انتقال پایدار ژن، یک گیاه مدل برای پژوهشهای علوم بیوتکنولوژی محسوب میگردد. ژن GDP-mannose-3´,5´-epimerase (GME) یکی از ژنهای کلیدیِ مسیر بیوسنتز ویتامین ث در گیاهان میباشد. در این تحقیق این ژن که از منبع کیوی جداسازی شده است به گیاه کاهو منتقل شد.
مواد و روشها: بهمنظور بهینهسازی کشت بافت کاهو آزمایشهایی جهت بررسی میزان کالوسزایی و باززایی غیرمستقیم با استفاده از اثرات نوع ریز نمونه (برگ لپهای و برگهای حقیقی) و 16 ترکیب تنظیم کننده رشد مختلف شامل غلظتهای 1/0، 02/0، 05/0 و 04/0 میلیگرم بر لیتر NAA و غلظتهای 1/0، 2/0، 4/0 و 6/0 میلیگرم بر لیتر BAP و جهت بررسی باززایی
غلظتهای 2/0، 4/0 و 6/0 میلیگرم بر لیتر BAP با سه تکرار به صورت آزمایش فاکتوریل در قالب طرحِ کاملاً تصادفی اجرا شدند. جهت انتقال ژن GME به این گیاه نیز آزمایشی با استفاده از رقم ستاره و سویه آگروباکتریوم (C58) روی دو نوع ریز نمونه (برگ لپهای و برگهای حقیقی) و با مدت زمان تلقیح دو و هشت دقیقه با سه تکرار به صورت فاکتوریل در قالب طرحِ کاملاً تصادفی انجام شد.
نتایج: نتایج حاصل از مقایسه میانگینها نشان داد که در ریز نمونههای برگ حقیقی و برگ لپهای غلظتهای 1/0 میلیگرم بر لیتر BAP و 04/0 میلیگرم بر لیتر NAA بیشترین کالوسزایی و باززایی غیرمستقیم را به میزان 100 درصد داشتند. نتایج حاصل از آزمایش انتقال ژن به کاهو حضور سازه موردنظر را در گیاهان تراریخته تأیید کرد.
نتیجهگیری: در آزمایش انتقال ژن، ریز نمونه برگهای حقیقی و مدت زمان تلقیح دو دقیقه با میزان 18 درصد تراریختی مناسبتر بودند.

تأثیر بیان ژن P19 بر افزایش میزان رونویسی و تولید پروتئین نوترکیب پلاسمینوژن بافتی انسانی (rtPA) در گیاه توتون (Nicotiana benthamiana)
هدف: فعالکننده پلاسمینوژن بافتی (rtPA) یکی از مهمترین داروها در درمان بیمارهای قلبی است. این دارو بهصورت پروتئین نوترکیب در سیستمهای بیانی تولید میشود که هزینههای تولید بسیار بالایی دارد. سیستم بیان موقت بهدلیل بیان زیاد، سرعت بالا، هزینه پایین و عدم تأثیرپذیری مکانی جهت بیان پروتئین بسیار مناسب میباشد. با این وجود مشخص شده است که خاموشی پس از رونویسی حاصل از کمپلکس RISC بر میزان بیان پروتئین نوترکیب تأثیر میگذارد. یکی از مهمترین سرکوب کنندههای خاموشی RNA شناخته شده در گیاهان، پروتئین P19 میباشد که از طریق میل ترکیبی زیادی که با siRNA دو رشتهای دارد به آن متصل میگردد و آن را تجزیه میکند و مانع خاموشی ژن میگردد. هدف از مطالعه حاضر بررسی تأثیر بیان همزمان ژن P19 بر بیان ژن فعالکننده پلاسمینوژن بافتی (rtPA) در سطح رونویسی و پروتئین در گیاه توتون Nicotiana benthamiana بود.
مواد و روشها: در تحقیق حاضر میزان بیان ژن rtPA در سطح رونویسی و پروتئین مورد بررسی قرار گرفت. در این تحقیق از تزریق همزمان اگروباکتریوم حاوی ناقل دوتایی pCAMBIA1304-rtPA و اگروباکتریوم حاوی ناقل pCAMBIA1304-P19 در مقایسه با اگروباکتریوم حاوی تنها ناقل بیانی pCAMBIA1304-rtPA استفاده شد. نمونههای برگی در روزهای 4، 7 و 10 روز پس از تلقیح با آگروباکتریوم تهیه شدند. سپس میزان رونویسی و پروتئین با استفاده از آزمون ReaTime PCR و الایزا محاسبه شد.
نتایج: نتایج آزمون Real Time PCR حاکی از افزایش 34 درصد میزان رونویسی ژن rtPA در حضور P19 نسبت به شاهد بود. بیشترین میزان رونویسی از ژنهای P19 و rtPA باگذشت چهار روز از تلقیح گیاهان با اگروباکتریوم حاصل شد. نتایج الایزا نشان داد که میزان بیان پروتئین rtPA در حضور ژن P19 در روز هفتم و دهم پس از تلقیح به ترتیب 89 و 84 میکروگرم بر گرم وزنتر برگ بود که در مقایسه با شاهد بهترتیب بهمیزان 12 و 15 درصد بیشتر بود.
نتیجهگیری کلی: نتایج نشان داد که کاربرد P19 علاوه بر سرکوب خاموشی ژن، میتواند برای دستیابی به بیان در سطح بالا مؤثر باشد.

بررسی الگوی بیان نسبی ژنهای کاندیدای تحمل به شوری در مرحله گیاهچهای برنج با استفاده از پیسیآر در زمان واقعی
هدف: با توجه به این که برنج منبع تغذیه بیش از نیمی از مردم جهان است. همچنین رشد و عملکرد این گیاه به شدت تحت تاثیر تنش شوری قرار میگیرد، تحقیق برای توسعه واریتههای متحمل به شوری امری ضروری است. لذا در این پژوهش، الگوی بیانی هفت ژن دخیل در تحمل به شوری با روش پیسیآر در زمان واقعی در ارقام برنج حساس (#6) ARC6578 و (#178) Shoemed و متحمل Bombilla (#48) مورد بررسی قرار گرفت.
مواد و روشها: نمونه RNA از گیاهچههای ۲۰ روزه برنج تحت تیمار NaCl (۱۰۰ میلیمولار) در سه زمان ۲۴، ۴۸ و ۷۲ ساعت پس از اعمال تنش شوری استخراج شد. بررسی بیان ژن بر روی هفت ژن کاندیدا (شامل ژنهای SOD, Cat1, 14-3-3 like protein GF14، Proxidase BP1 precursor، Zinc ion binding protein Plasma membrane H+-ATPase, و OsSIPK) با روش پیسیآر در زمان واقعی انجام شد. از ژن اکتین به عنوان ژن مرجع استفاده شد.
نتایج: بیان ژن OsSIPK در رقم متحمل، ۲۴ و ۴۸ ساعت بعد از تنش افزایش و ۷۲ ساعت بعد از تنش شوری کاهش یافت در حالی که بیان ژن Zinc ion binding protein در تمام ارقام و در همه ساعات پس از تنش کاهشی بود. بیان ژن PM H+-ATPase در ساعات اولیه پس از تنش در هر دو رقم حساس و متحمل افزایش یافت ولی با گذشت ۷۲ ساعت از تنش بیان این ژن در رقم حساس کاهش و در رقم متحمل افزایش یافت. بیان ژن 14-3-3 like protein GF14-6 ۴۸ ساعت پس از تنش در هر دو رقم حساس و متحمل به شدت افزایش یافت ولی با گذشت ۷۲ ساعت از تنش بیان این ژن در رقم حساس کاهش و در رقم متحمل افزایش یافت. بیان ژن Similar to Peroxidase BP1 precursor ۷۲ ساعت پس از تنش در رقم متحمل حدود ۴۰ برابر افزایش یافت. بیان ژن کاتالاز ۷۲ ساعت پس از تنش فقط در رقم متحمل افزایش معنیدار نشان داد. بیان ژن SOD از الگوی زمانی خاصی تبعیت نکرد هرچند بیان آن ۷۲ ساعت پس از تنش در رقم متحمل بیشتر از ارقام حساس بود.
نتیجهگیری: برخی ژنهای مورد مطالعه در این تحقیق با حذف گونههای فعال اکسیژن (ROS) در پاسخ عمومی گیاه به تنش شوری نقش دارند (مانند ژنهای SOD و CatA). از طرفی بین تحمل به شوری و میزان بیان برخی دیگر از ژنهای مورد مطالعه میتوان نوعی ارتباط وابسته به رقم-زمان را تشخیص داد (مانند ژنهای OsSIPK، PM H+-ATPase، 14-3-3 like protein GF14-6 و Peroxidase BP1).

شناسایی نشانگرهای ریزماهواره پیوسته با مقاومت به بیماری تغییر رنگ دانه در ژنوتیپهای برنج
هدف: بیماری تغییر رنگ دانه برنج به عنوان تهدید جدی بر عملکرد دانه و کیفیت آن در ارقام دیررس و بهنژادیشده در نواحی برنجکاری شمال ایران محسوب شده که موجب ایجاد خسارت در کیفیت و خصوصیات فیزیکی بذر میگردد. بدین جهت بهنژادی ژنوتیپهای برنج برای صفت مقاومت به بیماری، از استراتژیهای ترجیحی در مدیریت بیماری تغییر رنگ دانه در برنج محسوب میشود. یکی از مهمترین کاربردهای نشانگرهای مولکولی در برنامههای بهنژادی گیاهان، نقش آنها در افزایش کارایی بهنژادی سنتی گیاهان از طریق انتخاب غیرمستقیم با کاربرد نشانگرهای پیوسته با صفات هدف میباشد. از اینرو، در این پژوهش، از تجزیه ارتباطی برای شناسایی نشانگرهای پیوسته مرتبط با صفت مقاومت به بیماری تغییر رنگ دانه در ژنوتیپهای برنج استفاده شد.
مواد و روشها: در پژوهش حاضر، جهت ارزیابی فنوتیپی مقاومت 94 ژنوتیپ برنج شامل ارقام محلی و بهنژادیشده ایرانی و وارداتی به بیماری تغییر رنگ دانه طی دو سال زراعی 1396 و 1397 مورد بررسی قرار گرفت. ارزیابی ژنوتیپی با استفاده از128 نشانگر چندشکل ریزماهواره روی 94 ژنوتیپ برنج اجرا شد. سپس تجزیه ارتباطی از طریق مدل خطی عمومی (GLM) و مدل خطی مخلوط (MLM)، با استفاده از نرمافزار TASSEL انجام شد.
نتایج: نتایج ارزیابی فنوتیپی نشان داد که تنوع بالایی در ژنوتیپهای مورد مطالعه برای مقاومت به بیماری تغییر رنگ دانه وجود دارد. بر اساس تجزیه ساختار مبتنی بر مدل، ژنوتیپهای برنج مورد بررسی در دو زیرجمعیت کلاستربندی شدند. در تجزیه ارتباطی به دو روش GLM و MLM، بعد از تصحیح بنفرونی به ترتیب، 12 و 3 نشانگر ارتباط معنیداری را با صفت مقاومت به بیماری تغییر رنگ دانه در هر دو سال نشان دادند.
نتیجهگیری: نشانگرهای RM242، RM5709 و RM5955 در دو مدل آماری GLM و MLM ارتباط معنیداری با مقاومت به بیماری تغییر رنگ دانه در دو سال 1396 و 1397 نشان دادند. بنابراین میتوان از آنها به عنوان نشانگرهای پیوسته با صفت مقاومت در برنامههای بهنژادی برنج نظیر انتخاب به کمک نشانگر استفاده نمود.

ارزیابی تنوع ژنتیکی گندمهای اینکورن غرب ایران با استفاده از نشانگرهای ریزماهوارهای
هدف: وجود تنوع ژنتیکی در گیاهان زراعی و اجداد وحشی آنها نقش مهمی در پیشرفت برنامههای بهنژادی دارد. گونه اینکورن (Triticum monococcum L. ssp. boeoticum) در نواحی غربی ایران تنوع وسیع و پراکنش جغرافیایی گستردهای دارد. هدف از این تحقیق بررسی تنوع ژنتیکی گندمهای اینکورن جمعآوری شده از این منطقه با استفاده از نشانگرهای ریزماهوارهای بود. مواد و روشها:تنوع ژنتیکی 163 ژنوتیپ گندم اینکورن جمعآوری شده از 34 مکان متفاوت از سه استان غربی ایران با استفاده از 19 جایگاه ژنی SSR از ژنوم A گندم هگزاپلویید بررسی شد. نتایج: در 163 ژنوتیپ گندم اینکورن مورد مطالعه در مجموع 151 آلل چندشکل برای 19 جایگاه ریزماهوارهای با میانگین 94/7 شناسایی شد. دامنه محتوای اطلاعات چندشکلی (PIC) در ژنوتیپهای مورد مطالعه بین 64/0 برای جایگاه ژنی Xgwm480-3A تا 89/0 برای جایگاه ژنی Xgwm4-4A متغیر بود. میانگین محتوای اطلاعات چندشکلی در ژنوتیپهای مورد مطالعه 77/0 بود. با توجه به نتایج بدست آمده جایگاههای ژنیXgwm4-4A ، Xgwm610-4A و Xgwm282-7A مناسبترین جایگاهها برای بررسی تنوع ژنتیکی و تمایز ژنوتیپهای مورد مطالعه گندم اینکورن تشخیص داده شدند. میانگین ضریب شانون به میزان 16/0 نشان دهنده تنوع متوسط در ژنوتیپهای تحت بررسی بود. متوسط مکانهای ژنی چندشکلی در 34 جمعیت مورد مطالعه 74/36 درصد و میانگین هتروزیگوسیتی برابر 114/0 برآورد شد. بر اساس نتایج تجزیه واریانس مولکولی برای 34 جمعیت گندم اینکورن، اختلاف بین و درون جمعیتهای مورد مطالعه به ترتیب 7 و 93 درصد مشاهده شد. تجزیه خوشهای با استفاده از ضرایب جاکارد و بر اساس الگوریتم UPGMA ژنوتیپهای مورد مطالعه گندم اینکورن را در 10 گروه طبقهبندی نمود. طبق نتایج حاصل از تجزیه به مختصات اصلی دو بردار اولیه به ترتیب 33/29 و 01/24 درصد از کل واریانس ژنتیکی مولکولی را تبیین نمودند. نتیجهگیری: نتایج حکایت از مفید بودن نشانگرهای ریزماهواره در شناسایی و گروهبندی ژنوتیپهای گندم اینکورن داشت، بهطوریکه از اطلاعات بدست آمده میتوان در پروژههای اصلاحی، برنامهریزی حفاظت ژرم پلاسم و ایجاد کلکسیون از جمعیتهای گندم اینکورن استفاده نمود.

تجزیه و تحلیل ژنتیکی ناحیه 12S rRNA شترهای تککوهانه و دوکوهانه ایران
هدف: دانشمندان علوم دامی در سرتاسر جهان در پی یافتن منابع جدیدی از پروتئین حیوانی بوده و تلاش میکنند تا حیواناتی که بهترین ضریب تبدیل را دارند در اولویت کاری خود قرار دهند. از جمله این دامها میتوان به شتر اشاره کرد. تحقیقات جهت حفظ این ذخیره ژنتیکی با ارزش از حساسیت و اهمیت بیشتری برخوردار است. شناسایی تفاوتهای ژنتیکی در توالی ژنهای موجود در ژنوم میتوکندری با استفاده از ابزارهای بیوانفورماتیک، روشی سریع و قابل اطمینان برای شناسایی و طبقه بندی گونهها است و یکی از مؤثرترین ابزارها در برآورد تنوع زیستی ژنتیکی و روابط فیلوژنتیک در نژادهای مختلف دام از جمله شتر محسوب میشود. هدف از انجام این پژوهش توالییابی و مقایسه ساختار ژنتیکی ژن RNA غیر کدکننده زیر واحد کوچک ریبوزوم در شترهای تککوهانه و دوکوهانه ایران بود.
مواد و روشها: برای انجام این پژوهش از 20 نفر شتر تککوهانه و دوکوهانه ایرانی نمونه خون جمعآوری شد. پس از استخراج DNA ژن 12S rRNA با طول 1326 جفت باز در ژنوم میتوکندری توسط دو جفت آغازگر اختصاصی تکثیر و توالییابی شدند.
نتایج: وجود 3 هاپلوتایپ در شترهای دوکوهانه و 1 هاپلوتایپ در شترهای تککوهانه در جمعیت مورد پژوهش ثابت شد که این هاپلوتایپها به ترتیب دارای دو جایگاه چند شکل (SNP) در شتر دو کوهانه است و در بین شترهای تککوهانه هیچ تفاوت نوکلئوتیدی مشاهده نشد. درخت فیلوژنی پس از اخذ توالیهای مشابه ژن 12S rRNA میتوکندری دیگر گونههای موجود در بانک جهانی ژن با استفاده از آنها ترسیم شد.
نتیجهگیری: نتایج تجزیه و تحلیل فیلوژنی و تنوع ژنتیکی نشان داد گونههای شتر تککوهانه و دوکوهانه در دو گروه متفاوت قرار دارند و شترهای دوکوهانه ایرانی تنوع ژنتیکی بالاتری دارند که نشان از قدمت تکاملی طولانیتر این گونه دارد. اطلاعات در مورد تنوع ژنتیکی در میان نژادهای شتر میتواند توسعه برنامههای اصلاح نژاد را تسهیل کند و یک الزام برای حفظ منابع ژنتیکی است.
