ارزیابی مسیرهای بیوسنتز گاما آمینو بوتریک اسید (GABA) در نخود زراعی (Cicer arietinum L.) تحت تنش سرما
هدف: سازگاری متابولیکی گیاهان به تنش سرما نقش مهمی در رشد، بقا و عملکرد گیاهان زراعی دارد. گاماآمینوبوتریک اسید (GABA) به عنوان اسمولیت احتمالا در جهت مقابله با تنش اکسیداتیو القا شده توسط سرما مشارکت دارد. روش: در این پژوهش میزان GABA، پوتریسین(Put)، پراکسیدهیدروژن (H2O2)، فعالیت آنزیم دیآمیناکسیداز (DAO) و بیان نسبی ژن گلوتامات دکربوکسیلاز (GAD1) (موثر در بیوسنتز GABA) در دو ژنوتیپ متحمل (Sel96th11439) و حساس (ILC533) نخود زراعی (Cicer arietinum L.) تحت تنش سرما چهار درجه سلسیوس به صورت آزمایش فاکتوریل در قالب طرح کاملا تصادفی مطالعه شد. نتایج:در ژنوتیپ متحمل میزان H2O2 پس از افزایش معنیدار در روز اول تنش، در روز ششم کاهش معنیداری در مقایسه با شاهد نشان داد (بیش از 7/4 درصد در حالیکه تجمع آن در ژنوتیپ حساس مشاهده شد (تا50 درصد)، نتایجی که بیانگر سازگاری نسبی به سرما در ژنوتیپ متحمل بود. تحت تنش سرما میزان متابولیتهایGABA و Putدر ژنوتیپ متحمل به ترتیب تا 14 و 35 درصد بیشتر از ژنوتیپ حساس بود. بهموازات افزایش میزان GABA تحتتنشسرما، در ژنوتیپ متحمل فعالیت آنزیم DAO و بیان نسبی ژن GAD1بهعنوان مسیرهای بیوسنتز این متابولیت افزایش معنیدارییافت (به ترتیب تا 3 و 17 برابر). حداکثر فعالیت این دو مسیر در ژنوتیپمتحملدرروزششم پسازتنشسرمامشاهدهشد. نتیجهگیری: تحت تنش سرما تجمع GABA در ژنوتیپ متحمل منجر به کاهش آسیب سلولی (نتایج H2O2) و بهبود درجه تحمل نخود به سرما شد. چنینشاخصهاییدرارزیابیژنوتیپهاینخودتحتتنش سرما موثر بوده و بکارگیریآنهادربرنامههایبهنژادیمفیداست.

تاثیر گزینش ژن Ppd-D1a و زودسنبلهدهی بر صفات مهم زراعی گندم نان (Triticum aestivum L.)
هدف: ژنهای کنترلکننده فتوپریود (Ppd) مهمترین گروه ژنی کنترلکننده زودرسی در گندم نان هستند. هدف این پژوهش بررسی این ژنها در ارقام ایرانی و بررسی نقش Ppd-D1a در زودرسی بود. مواد و روشها: زودرسی با استفاده از تلاقی برگشتی از اکسکلیبر به رقمهای کلحیدری، روشن و مهدوی منتقل و نسل BC1F3 ایجاد شد. ژنهای کنترلکننده فتوپریود Ppd-A1، Ppd-B1 و Ppd-D1 در ارقام ایرانی با استفاده از آغازگرهای اختصاصی بررسی شد. با استفاده از آنالیز تفرق بالک مکان ژنی Ppd-D1 در نتاج زودرس سه جمعیت بررسی گردید. با ارزیابی دوساله در مزرعه، تاثیر گزینش برای زودرسی بر صفات مهم زراعی گندم نان بررسی شد. نتایج: ارقام مهدوی، قدس، کلحیدری، سرداری، روشن، کویر، اکسکلیبر و شاهپسند داری ژنهای Ppd-A1b و Ppd-B1b بودند. کلحیدری، سرداری، روشن و کویر آلل Ppd-D1b و مهدوی، قدس، اکسکلیبر و شاهپسند آلل Ppd-D1a را دارا بودند. ارزیابی بالک 10% از زودسنبلهدهترین نتاج نسل BC1F3 جمعیتها نشان داد همه نتاج زودرس دارای ژن Ppd-D1a بودند. زودسنبلهدهی و زودرسی همبستگی مثبت و معنیداری داشتند. با این وجود، تعداد روز تا سنبلهدهی با طول دوره پرشدن دانه همبستگی منفی و معنیداری داشت، که باعث افزایش وزن هزار دانه، تعداد دانه در بوته و عملکرد دانه شد. گزینش برای زودسنبلهدهی باعث افزایش ارتفاع بوته، اندازه برگ، تعداد سنبله در بوته، تعداد دانه در بوته، وزن هزار دانه و عملکرد دانه و کاهش طول سنبله، طول ریشک و تعداد پنجههای نابارور شد. نتیجه گیری: گزینش به کمک نشانگر و یا تلاقی برگشتی به کمک نشانگر برای ژن Ppd-D1a میتواند باعث زودرسی گندم نان شود. گزینش برای زودسنبلهدهی باعث بهبود این صفت در زمینه ژنتیکی روشن، کلحیدری و مهدوی به ترتیب به مدت 13، 14 و 10 روز شد. در شرایط کرمان، گزینش برای زودرسی باعث بهبود اکثر صفات مهم تاثیرگذار بر عملکرد شد.

تجزیه و تحلیل ژنتیکی و فیلوژنتیکی ناحیه D-loop ژنوم میتوکندری بز نجدی
هدف: تجزیه و تحلیل DNA میتوکندری به دلیل ویژگیهایی مانند تعداد کپی بالا، عدم نوترکیبی، نرخ جایگزینی بالا و وراثت مادری، یک ابزار قدرتمند در درک ما از تکامل بوده است. هدف از این مطالعه تجزیه و تحلیل ژنتیکی و فیلوژنتیکی ناحیه D-loop ژنوم میتوکندری در بز نژاد نجدی و همچنین بررسی ارتباط این نژاد با سایر نژادهای موجود در ایران و جهان بود. مواد و روشها: 12 نمونه خون بز نجدی غیرخویشاوند از نقاط مختلف استان خوزستان تهیه شد. پس از استخراج DNA یک قطعه 968 جفت نوکلئوتیدی ناحیه D-loop ژنوم میتوکندری تکثیر و توالییابی شد. سپس با توالی نوکلئوتیدی نژادهای دیگر ثبت شده در پایگاه NCBI مورد مقایسه قرار گرفت. در نهایت درخت فیلوژنتیک با استفاده از نرمافزار MEGA6 ترسیم گردید. نتایج: نتایج نشان داد که نژاد بز نجدی در هاپلوگروپ A قرار میگیرد و با نژادهای بز چینی و پاکستانی کمترین فاصله ژنتیکی را دارد. نتایج آنالیز تنوع ژنتیکی در جمعیت بز نجدی منجر به شناسایی 20 جهش با 5 هاپلوتیپ مختلف گردید. همچنین میزان تنوع هاپلوتیپی، نوکلئوتیدی و تعداد متوسط نوکلئوتیدهای مختلف (Tajima's D) به ترتیب 727/0، 009/0 و 54/0 برآورد شد که نشاندهنده تنوع بالا در این نژاد است. محاسبه شاخص تاجیما D نشان میدهد که در جمعیت بز نجدی، انتخاب طبیعی متعادل در حال انجام است و فراوانی آللهای کمیاب درون این جمعیتها کم است. نتیجهگیری: تنوع ژنتیکی بز نجدی در طی سالهای متمادی افزایش یافته است و این در حالی است که تعداد این دام در استان خوزستان کاهش یافته است. افزایش تنوع ژنتیکی میتواند به دلیل آمیختهگری این نژاد با نژادهای پاکستانی و غیره باشد. نتیجه این تلاقیگریها در آیندهای نزدیک انقراض بز نجدی خواهد بود و این موضوع توجه بیشتر مسئولان را میطلبد.

تأثیر ترکیبات محیط کشت بر القای ریشههای مویین در کاسنی (Cichorium intybus L.) و افزایش متابولیتهای ثانویه
هدف: کاسنی گیاه دارویی مهمی از خانوادهی گلستارهایها ( (Asteraceaeبوده و دارای ترکیبات دارویی ارزشمندی میباشد. القای ریشههای مویین با استفاده از rhizogenes Agrobacterium یکی از روشهای کاربردی برای افزایش بیوسنتز متابولیتهای ثانویه میباشد.
مواد و روشها: در این تحقیق، در آزمایش اول، تأثیر غلظتهای مختلف عناصر ماکرو محیط کشت بر کارایی القای ریشههای مویین در کاسنی بررسی شد. تائید مولکولی ریشههای مویین به وسیلهی PCR با استفاده از آغازگرهای اختصاصی ژن rolB انجام شد. در آزمایش دوم میزان رشد لاینهای مختلف ریشههای مویین کاسنی بررسی شد و همچنین تاثیر الیسیتور هورمونی و منبع کربن و بر رشد ریشههای مویین بررسی شد.
نتایج: نتایج بهدست آمده نشان داد که بیشترین درصد القای ریشههای مویین در غلظت x1 پتاسیم نیترات، x5/1 نیترات آمونیوم، x1 و x5/1 منیزیم سولفات، x5/1 کلسیم کلرید و x5/0 پتاسیم فسفات (بهترتیب 66/46، 33/53، 66/46، 33/53 و 66/66 درصد) مشاهده شد. بیشترین میزان فنول کل در غلظت x1 پتاسیم نیترات، x2 نیترات آمونیوم، x5/1 منیزیم سولفات، x5/1 کلسیم کلرید و x5/0 پتاسیم فسفات (بهترتیب 21/4، 33/4، 26/4، 58/4 و 84/4 میلیگرم بر گرم وزن خشک) بهدست آمد. غلظت x5/1 پتاسیم نیترات، x2 نیترات آمونیوم، x1 منیزیم سولفات، x5/1 کلسیم کلرید و x5/0 پتاسیم فسفات بالاترین میزان فلاونوئید را داشتند (بهترتیب 25/17، 52/18، 22/17، 3/18 و 96/17 میلیگرم بر گرم وزن خشک). لاین C بیشترین افزایش زیستتوده (36/0 گرم) را نشان دادند همچنین بیشترین وزن تر (96/1 و 72/1 گرم در فلاسک) به ترتیب در ریشههای رشد کرده در محیط کشت حاوی 5/1 میلیگرم در لیتر NAA و 3 درصد و 4 درصد ساکارز بهدست آمد.
نتیجهگیری: کشت ریشههای مویین یک فرآیند نوین برای تولید متابولیتهای ثانویه در سطح وسیع است که قابل استفاده در صنایع داروسازی و غذایی میباشد. نتایج این پژوهش نشان داد که استفاده از سویهی ATCC15834 و محیط همکشتی حاوی x 5/0 KH2PO4 باعث افزایش میزان القای ریشههای مویین و متابولیتهای گیاه کاسنی شد.

تجزیه و تحلیل مجموعههای ژنی جهت شناسایی ژنها و مسیرهای زیستی مرتبط با صفات وزن تخممرغ در کل دوره تولید
هدف: شناسایی ژنهای بزرگ اثر مؤثر بر صفات مهم اقتصادی یکی از مهمترین اهداف اصلاح نژادی در پرورش مرغ است. پژوهش حاضر به منظور مطالعه پویش ژنومی بر اساس آنالیز مجموعههای ژنی برای شناسایی جایگاههای ژنی مؤثر بر صفات مرتبط با وزن تخممرغ نژاد رُد آیلند رِد با استفاده از آرایههای ژنومی با تراکم بالا بوده است.
مواد و روشها: اطلاعات رکوردهای فنوتیپی و ژنوتیپی مرتبط با وزن تخممرغ نمونهها از پایگاه ذخیره ژنومی برخط Figshare استفاده شد. آنالیز پویش کل ژنومی بر پایه آنالیز مسیر در سه مرحله تعیین مکان SNPهای معنیدار با ژن، ارتباط ژنها به طبقات عملکردی و مسیرهای بیوشیمیایی و پویش کل ژنومی بر پایه آنالیز مسیر انجام میشود. بر این اساس، مطالعه پویش ژنومی برای صفات وزن تخممرغ در اولین تخمگذاری و 28، 36، 56، 66، 72 و 80 هفتگی در 1078 قطعه مرغ در برنامه GenABEL انجام شد. سپس با استفاده از بسته نرم افزاری biomaRt2 ژنهای معنیداری که در داخل و یا 20 کیلوباز بالادست یا پایین دست نشانگرهای SNP معنیدار قرار داشتند، شناسایی گردید. در نهایت تفسیر مجموعه ژنی با بسته نرم افزاری goseq برنامه R با هدف شناسایی عملکرد بیولوژیکی ژنهای نزدیک به مناطق انتخابی و ژنهای کاندیدا از طریق پایگاههای GO، KEGG، DAVID و PANTHER انجام شد.
نتایج: در این پژوهش تعداد 9 نشانگر تک نوکلئوتیدی واقع روی کروموزومهای 3، 4، 6، 7، 8، 9، 19، 20 و 22 شناسایی شدند که با ژنهایMC3R ،LEPR ، ECT2، SH3GL2، KCNMA1، SPP1، PCK1، MMP9، PPP1CB، ACOX1 و IGFBP2 مرتبط بودند. تعدادی از این نشانگرهای شناسایی شده در مناطق ژنومی معنیدار با مطالعه پیشین مرتبط با وزن تخممرغ همخوانی داشتند. در تفسیر مجموعه ژنی تعداد 28 مسیر هستی شناسی ژنی و بیوشیمیایی با صفات وزن تخممرغ شناسایی شد (P˂0.01). از این بین، مسیرهایRegulation of feeding behavior،Positive Regulation of apoptotic process،Positive regulation of protein phosphorylation osteoblast differentiation، Positive regulation of gluconeogenesis، Cell-cell junction وFocal adhesion عملکردهای مهمی را در ارتباط با وزن تخممرغ و فرآیند تولید تخم مرغ از طریق رشد اووسیت و تخمکاندازی، تشکیل پروتئین آلبومین و تشکیل پوسته تخم مرغ بر عهده داشتند.
نتیجه گیری: با توجه به تأیید نتایج حاصل از مطالعه قبلی در زمینه پویش ژنومی صفات وزن تخممرغ و شناسایی مناطق ژنومی جدید استفاده از یافتههای این تحقیق میتواند در انتخاب ژنتیکی برنامههای اصلاح نژادی مرغ مفید باشد.

بررسی تنوع ژنتیکی برخی از ژنوتیپهای پونه وحشی (Mentha longifolia L.) ایران با استفاده از نشانگر ISSR و ارتباط آن با عملکرد ماده خشک و درصد اسانس
هدف: هدف از این پژوهش بررسی تنوع ژنتیکی ژنوتیپهای مختلف پونه وحشی در شرایط کشت یکسان است که میتواند بهعنوان مقدمهای بر اهلیسازی، حفظ ژرمپلاسم و امکان تلاقی بین ژنوتیپهای آن جهت برنامههای بهنژادی آینده باشد.
مواد و روشها: در این پژوهش پس از تهیه 20 ژنوتیپ مختلف از سراسر ایران، کشت آنها در قالب طرح بلوکهای کامل تصادفی در سه تکرار تحت شرایط یکسان انجام شد. پس از استخراج DNA بررسی تنوع ژنتیکی ژنوتیپها با استفاده از 12 نشانگر ISSR از بین 15 نشانگر توسط واکنش زنجیرهای پلیمراز (PCR) انجام گرفت. استخراج اسانس برای هر ژنوتیپ به روش تقطیر با آب انجام شد. عملکرد ماده خشک برحسب گرم بر متر مربع و میزان اسانس بهصورت وزنی-وزنی، براساس وزن خشک اندازهگیری شد. همچنین، ارتباط نشانگرهای مولکولی با صفات عملکرد ماده خشک و بازده اسانس با استفاده از رگرسیون گام به گام تعیین گردید.
نتایج: میانگین درصد چندشکلی تعیین شده در مجموع ژنوتیپهای مورد بررسی 97/91 بود. تعداد باندهای چندشکل هر آغازگر از 5 تا 9 عدد متغیر بود و در مجموع 89 باند تکثیری امتیازدهی شدند که از این تعداد 82 مکان، چندشکلی نشان دادند. میانگین محتوای اطلاعات چندشکلی آغازگرها (PIC) 31/0 برآورد گردید و آغازگر IS1 بالاترین مقدار PIC (46/0) را نشان داد. همچنین شاخص نی و شانون برای آغازگر IS1 بهترتیب 43/0 و 61/0 بود. بررسی دندروگرام حاصل از تجزیه خوشهای با استفاده از ضریب تشابه جاکارد و الگوریتم UPGMA ژنوتیپهای مورد بررسی را در چهار گروه قرار داد. بیشترین فاصله ژنتیکی بین ژنوتیپهای خوزستان و قزوین با ضریب 39/0 و کمترین فاصله بین ژنوتیپهای کرمان-2 و کرمان-4 با ضریب تشابه 77/0 مشاهده شد. رگرسیون گام به گام نشان داد که نشانگر IS10 با ضریب تبیین حدود 70/0 بیشترین همبستگی را با صفات درصد اسانس و عملکرد ماده خشک دارد.
نتیجهگیری: نتایج این پژوهش نشان داد که نشانگرهای ISSR بهطور مؤثری میتوانند برای مطالعه تنوع ژنتیکی ژنوتیپهای پونه وحشی استفاده شوند. آغازگرهای IS1 و IS10 با داشتن بهترین شاخصهای نشانگری بهعنوان بهترین آغازگرها معرفی شدند. همچنین دو ژنوتیپ خوزستان و قزوین بیشترین فاصله ژنتیکی را داشتند.

بررسی حضور آنزیمهای خارج سلولی در کشت قارچهای اندوفیت جدا شده از دو گیاه فرفیون و سلمه تره
مقدمه: آنزیمها، متابولیتهای با ارزش میکروبی هستند که در صنایع مختلف کاربرد وسیعی دارند. در حال حاضر تولید آنزیمهای خارج سلولی توسط میکروارگانیسمها، یکی از بهترین و موثرترین روشها به منظور تولید صنعتی این آنزیمها است. آنزیمهای هیدرولازی از مهمترین این آنزیمهای خارج سلولی هستند. که توسط میکروارگانیسمهای مختلف از جمله قارچهای اندوفیتها، تولید می گردند.
مواد و روشها: به منظور بررسی اندوفیتهای دو گیاه فرفیون و سلمهتره، در تابستان1395، از شش بافت مختلف گل، برگ، بذر، ریشه، طوقه و ساقه ی گیاهان سالم دو منطقه استان همدان نمونه برداری صورت گرفت. پس از جدا و خالص سازی، قارچهای اندوفیت حضور آنزیمهای هیدرولازی خارج سلولی آمیلاز، کاتالاز، پروتئاز، کازئیناز، سلولاز، پکتیناز و زایلاناز با اضافه کردن سوبسترای مربوطه در محیط کشت قارچ بررسی گردید.
نتایج: پس از استریل سطحی و انتقال نمونهها به محیط کشت پوتیتو دکستروز آگار، مجموعاً از دو گیاه فرفیون و سلمه تره، 31 جدایه قارچ اندوفیت جداسازی شد. نتایج نشان داد: 84 درصد از جدایههای قارچی دارای فعالیت آنزیمهای خارج سلولی آمیلاز، 81 درصد کاتالاز، 71 درصد فعالیت پروتئاز، 61 درصد سلولاز، 39 درصد پکتیناز و 26 درصد زایلاناز بودند. بررسیهای مورفولوژیکی قارچهای جدا شده نشان داد که برخی از این قارچها متعلق به جنسهای فوزاریوم، آلترناریا و تریکودرما بودند.
نتیجهگیری: نتایج بدست آمده در این تحقیق نشان داد که قارچهای اندوفیت جدا شده ازدو گیاه سلمه تره و فرفیون قادر به تولید انواع آنزیم های هیدرولیزی در شرایط آزمایشگاهی را داشتند. در این میان، حضور آنزیم های مهمی مانند سلولاز، پکتیناز و پروتئاز در تعدادی از جدایه ها، اهمیت نتایج این تحقیق را بیشتر میکند. با توجه به کاربرد وسیع این آنزیمها و اهمیتی که در صنایع مختلف دارند ، اهمیت این قارچها و نیاز به بررسی بیشتر بر روی عملکرد آنزیمی آنها مشخص می گردد.

شناسایی تعدادی از ژنهای با الگوی بیان متفاوت بین ارقام مقاوم و حساس گندم در پاسخ به تنش خشکی با استفاده از روش متاآنالیز
هدف:گندم از مهمترین منابع غذایی بشر میباشد که در تولید این محصول، تنشهای غیرزیستی مانند خشکی، شوری، دما و غیره دخیل میباشند. از این رو شناسایی و ارزیابی ژنهای دخیل در مقاومت به تنشها در این گیاه از اهمیت بالایی برخوردار است. این تحقیق با هدف شناسایی ژنهای پاسخ دهنده به خشکی و بررسی تفاوت بیان آنها در گندم حساس و مقاوم به خشکی، انجام شد. مواد و روشها: از بین دادههای موجود در سایت NCBI GEO در سال 2018 دادههای ریزآرایه مورد نظر جمعآوری شدند. کتابخانههای تهیه شده دارای ارقام مقاوم و حساس به خشکی، تحت دو شرایط تنش و کنترل بودند. با استفاده از متاآنالیز، ژنهای تغییر بیانیافته تحت شرایط تنش برای هر گروه از ارقام شناسایی شد. از ژنهای تغییر بیان یافته معنیدار شناسایی شده، ژن دئوکسی ژنز کاهشدهنده شبهپروتئین aci (ARD) برای تایید آزمایشگاهی مورد ارزیابی قرار گرفت. نمونهبرداری از برگها در مرحله 4 برگی و در سه زمان 2، 4 و 7 روز بعد از اعمال تنش همراه با نمونهی کنترل، با سه تکرار بیولوژیکی انجام گرفت. استخراج RNA از نمونههای برگی با استفاده از محلول استخراج RNx™-plus و واکنش Real time PCR با استفاده از آغازگرهای اختصاصی ژن ARD انجام گرفت. نتایج: در بین پروبهای تغییر بیانیافته، به ترتیب 658 و 1561پروب ایدی به طور اختصاصی در ارقام مقاوم و حساس، در شرایط تنش افزایش بیان داشتند. ژنهای فاکتورهای رونویسی Myb3، ethylene responsive 5a، MIKC-type MADS-boxWM24B و salinity inducible ERF4 در ارقام مقاوم و فاکتورهای رونویسی WRKY15 ، MADS-box TaAGL8، WRKY39 و Myb در ارقام حساس دارای افزایش بیان معنیداری بودند. با کمک متاآنالیز ژنها و فاکتورهای رونویسی، با بیان متفاوت بین ارقام حساس و مقاوم گندم شناسایی شدند و از این نتایج، میتوان برای شناسایی ارقام مقاوم و حساس گندم نسبت به تنش خشکی استفاده کرد. نتایج Real time PCR ژن ARD برای 2 رقم گندم مقاوم به خشکی 6o4 و رقم حساس به تنش خشکی Sundor همسو با نتایج متاآنالیز انجام شده بود و نتیجهی متاآنالیز را تایید کرد. نتیجهگیری: با توجه به بررسی رفتار بیانی این ژن و دیگر نتایج بدست آمده متاآنالیز در این مطالعه ، میتوان گامی موثر در پیشبینی مقاومت و حساسیت به خشکی در ارقام مختلف گندم برداشت.

تولید ایمنوگلوبین Y در زرده تخم مرغ علیه باکتری های سالمونلا تیفیموریوم و انتریتیدیس
هدف: سالمونلا یک باکتری گرم منفی، بیهوازی اختیاری و تاژکدار است که در جنس سالمونلا و در خانواده انتروباکتریاسه قرار دارد که به عنوان یک عامل بیماری عفونی مشترک در انسان و حیوانات شناخته شده است. در سالهای اخیر، انواع مختلفی از آنتی بیوتیکها برای مقابله با عفونت سالمونلا استفاده شده است که با افزایش تعداد باکتریهای مقاوم به آنتی بیوتیک، استفاده از روشهای جایگزین ضروری به نظر میرسد. ایمونوگلوبین Y (IgY) اختصاصی زرده تخممرغ باعث ایجاد ایمنی غیرفعال در جنین میشود و استخراج این آنتیبادیها از زرده تخممرغ در مقایسه با سایر آنتیبادیها ارزانتر و آسانتر هستند. مطالعات زیادی نشان دادند که IgY زرده تخممرغ دارای عملکرد ایمنیزایی در تعداد زیادی از عفونتهای باکتریایی و ویروسی در پستانداران هستند. هدف از انجام این مطالعه تولید IgY اختصاصی علیه باکتریهای سالمونلا تیفیموریوم و انتریتیدیس در زرده تخممرغ مرغهای تخمگذار بود.
مواد و روش ها: بدین منظور باکتریهای کشته شدهی سالمونلا تیفیموریوم و انتریتیدیس به میزان یک میلیلیتر در عضله سینه شش قطعه مرغ تخمگذار نژاد لگهورن سفید در سن 11 هفتگی تزریق شدند. تزریقها هر دو هفته تکرار شده و یک هفته بعد از آخرین تزریق تخممرغها جمع آوری و IgY زرده تخممرغ با استفاده از روش پولسون و پلیاتیلنگلیکول 6000 استخراج شد. وجودIgYهای تولید شده به کمک PAGE - SDS تایید و وجود IgY های اختصاصی با انجام آزمون الایزای غیرمستقیم اختصاصی با استفاده از باکتریهای کشتهشده سالمونلا تیفیموریوم و سالمونلا انتریتیدیس تایید شد.
نتایج: نتایج PAGE-SDS حضور باندهای پروتئینی 27 و 67 کیلودالتون که مربوط به زنجیرههای سبک و سنگین IgY هستند را تایید کرد و آزمون الایزای غیرمستقیم اختصاصی نشاندهنده تولید آنتیبادی اختصاصی علیه سالمونلا تیفی موریوم و انتریتیدیس در زرده تخممرغ بود.
نتیجهگیری: ممکن است بتوان از زرده تخم مرغهای حاوی IgY اختصاصی برای سالمونلا تیفیموریوم و انتریتیدیس، با ایجاد ایمنی غیرفعال جهت جلوگیری از برخی از بیماریهای گوساله مثل اسهال استفاده کرد.

پروفایل بیانی mRNA مختص بافت ژن BMP15 در بز
هدف: سه ژن باروری BMPR1B (یا ALK6 معروف به FecB روی کروموزوم 6)، GDF9 (معروف به FecG روی کروموزوم 5) و BMP15 (معروف به FecX روی کروموزوم X) شناسایی شده است. ژن های پروتئین مورفوژنتیک استخوان (BMP) اعضای فوق خانواده فاکتور رشد تبدیل کننده بتا (TGF-β) هستند که سیتوکین های چند منظوره با عملکرد 2 برابر هستند و در انواع سلولها بیان میشوند. BMP ها در اصل بر اساس توانایی آنها در تولید استخوان در جای غیر معمول هنگام کاشت در داخل بافت نرم در داخل بدن شناسایی میشوند. آنها همچنین در رشد و توسعه جنین، هموستازی، ترمیم الگوسازی بافتهای مختلف، تمایز سلولی و آپوپتوز نقش دارند. ژن BMP15 با افزایش میتوز در سلول گرانولوزا، سرکوب بیان گیرنده هورمون محرک فولیکول و تحریک بیان لیگاند کیت، تکثیر و تمایز سلول گرانولوز را تنظیم میکند. از این رو، این ژن نقشی اساسی در باروری پستانداران ماده دارد. هدف این مطالعه پروفایل بیانی mRNA مختص بافت ژن BMP15 در بز کرکی راینی با استفاده از Real Time PCR بود.
مواد و روشها: نمونههای بافتی (از هر بافت 3 تکرار) از تخمدان، رحم، شاخ رحم، اویداکت، ماهیچه اسکلتی، چربی زیرپوستی، قلب، جگر، شش، کلیه، طحال و غده فوق کلیه در هنگام کشتار در کشتارگاه از دو راس بز تهیه شد. استخراج RNA کل و سنتز cDNA انجام شد شد. میزان نسبی بیان ژن BMP15 با استفاده از واکنش Real Time PCR به روش SYBR Green به دست آمد. ژن GAPDH به عنوان ژن کنترل داخلی استفاده شد. برای تجزیه و تحلیل دادههای حاصل از Real Time PCR از روش پیفافل استفاده شد.
نتایج: بیان ژن BMP15 در بافتهای تخمدان، رحم، شاخ رحم، اویداکت، ماهیچه اسکلتی، چربی زیرپوستی، قلب، جگر، شش، کلیه، طحال و غده فوق کلیه بز کرکی راینی با استفاده از روش PCR در زمان واقعی نشان داد که این ژن فقط در تخمدان بیان میشود و در بافتهای مطالعه شده دیگر بیان نمیشود.
نتیجهگیری: با توجه به نتایج پژوهش حاضر و نتایج سایر محققین میتوان نتیجه گرفت که ژن BMP15 نقش مهمی در تخمدان و در نتیجه در باروری دارد، چرا که در پژوهش حاضر مشخص شد که ژن BMP15 فقط در بافت تخمدان بیان میشود. لذا، ژن BMP15 به احتمال زیاد برای باروری مادهها بسیار ضروری است و نتایج این پژوهش اساسی را برای پژوهشهای آینده جهت توصیف نقش ژن BMP15 به عنوان یک ژن کاندیدا برای باروری بهتر و فیزیولوژی طبیعی در حیوانات اهلی، به ویژه بز فراهم کرده است.
