بیان ژن های باروری در بافت بیضه خروس‌های مسن مادرگوشتی تحت تاثیر اسید آمینه سرین و مکمل کولین جیره

چکیده

هدف: با افزایش سن، استرس اکسیداتیو و آسیب به غشای اسپرم باعث کاهش کیفیت، تحرک و باروری خروس‌ها می‌شود. تغییراتی مانند کاهش فسفاتیدیل کولین و افزایش فسفاتیدیل سرین ساختار غشا و کیفیت اسپرم را تحت تأثیر منفی قرار می‌دهد. افزودن مکمل‌های سرین و کولین به رژیم غذایی، با تقویت مقاومت غشای اسپرم در برابر استرس اکسیداتیو، می‌تواند سلامت و باروری خروس‌های مسن را بهبود دهد. ژن‌های PVRL3 و StAR نیز با نقش کلیدی در رشد، عملکرد اسپرم و سنتز هورمون‌های جنسی، هدف پژوهش حاضر برای بررسی تأثیر این مکمل‌ها در تقویت باروری هستند.
مواد و روش‌ها: این مطالعه روی 36 خروس مادر گوشتی مسن نژاد راس 308 بمدت 10 هفته انجام شد. خروس‌ها بصورت تصادفی در 6 تیمار مختلف قرار گرفتند. تیمارها شامل: 1) جیره پایه حاوی صفر درصد اسید آمینه سرین+ صفر درصد کولین، 2) صفر درصد سرین + تامین 470 میلی‌گرم بر کیلوگرم کولین، 3) 15/ 0 درصد اسیدآمینه سرین + صفر درصد مکمل کولین، 4) 15/ 0 درصد اسید آمینه سرین + تامین 470 میلی‌گرم بر کیلوگرم کولین، 5) 3/ 0 درصد اسیدآمینه سرین + جیره حاوی صفر درصد مکمل کولین و 6 ) 3/ 0 درصد اسید آمینه سرین + تامین 470میلی‌گرم کولین بودند. در انتهای آزمایش، 4 خروس از هر تیمار کشتار و قسمتی از بافت بیضه جدا شده و در دمای 80- ذخیره گردیدند. بیان نسبی ژن‌های StAR و PVRL3 با روش Real-time PCR اندازه‌گیری و با نرمالسازی نسبت به ژن مرجع (β-actin) تحلیل شدند.
یافته‌ها: نتایج حاصل از این پژوهش نشان داد، افزودن سرین در جیره خروس‌ها باعث کاهش معنی‌دار بیان ژن PVRL3 و افزایش معنی‌دار بیان ژن StAR شد (P<0.05) و افزودن کولین نیز بیان هر دو ژن را بطور معنی‌داری افزایش داد (P<0.05). اثر متقابل سرین و کولین بر بیان ژن StAR معنی‌دار نبود (P>0.05) اما همزمانی افزودن این دو به جیره منجر به افزایش معنی‌دار بیان این ژن شد (P<0.05). افزودن همزمان 15/0 و 3/0 درصد اسید آمینه سرین به همراه 470 میلی‌گرم بر کیلوگرم کولین سبب کاهش معنی‌دار بیان ژن PVRL3 شده (P<0.05) که نشان می‌دهد، حضور کولین نتوانسته از کاهش بیان ژن PVRL3 توسط سرین جلوگیری کند.
نتیجه‌گیری: مصرف مکمل کولین و سرین باعث افزایش قابل‌توجه باروری در خروس‌های مسن می‌شود. کولین با تقویت ساختار غشای اسپرم و افزایش انرژی میتوکندری و سرین نیز با سنتز گلوتاتیون، از اسپرم در برابر استرس اکسیداتیو محافظت می‌کنند و منجر به بهبود عملکرد بیضه‌ها شده و باروری را بهبود می‌بخشند.

چکیده [English]

Objective: Oxidative and membrane damage reduce the fertility, motility, and quality of the sperm with increasing age. Changes such as lowered phosphatidylcholine and increased phosphatidylserine weaken the membrane structure and the health of the sperm. Supplementation with choline and serine in the diet can increase the resistance of the sperm membrane to oxidative damage and increase the fertility and health of aging roosters. The StAR and PVRL3 genes, which are critical in sperm development, function, and sex hormone production, are among the genes targeted by this research to establish the effect these supplements have on the improvement of fertility.
Materials and methods: This Study was conducted on 36 aged Ross 308 breeder cockerels for 10 weeks. The cockerels were randomly allocated into six treatment groups: 1) basal diet containing 0% serine and 0% choline, 2) 0% serine and 470 mg/kg supplementation of choline, 3) 0.15% serine without choline supplementation, 4) 0.15% serine with 470 mg/kg supplementation of choline, 5) 0.3% serine without choline supplementation, and 6) 0.3% serine with 470 mg/kg supplementation of choline. Four cocks from each group were killed at the end of the trial, and testicular tissue samples were collected and preserved in –80°C. The relative expression of the StAR and PVRL3 genes was identified using real-time PCR and was compared with the reference gene β-actin after normalization.
Results: The results of this study showed that serine supplementation in the diet of roosters greatly reduced PVRL3 gene expression and greatly enhanced the StAR gene expression (P<0.05). Supplementation with choline elevated expression of both genes greatly (P<0.05). Although the effect of serine and choline in combination on the expression of the StAR gene was not significant (P>0.05), combined supplementation always had a significant enhancement of StAR expression (P<0.05). Additionally, simultaneous addition of 0.15% and 0.3% serine along with 470 mg/kg choline caused a significant decrease in PVRL3 expression (P<0.05), indicating that choline could not prevent the serine-induced reduction in PVRL3 gene expression.
Conclusions: Supplementation with choline and serine significantly enhances fertility in aging cocks. Choline increases sperm membrane structure and mitochondrial ATP yield, and serine sustains glutathione production that keeps sperm away from oxidative stress. Both enhance testicular function and overall reproductive performance.
