اثر رازیانه (Foeniculum vulgare) بر بیان ژن فاکتور 1 رشد شبه انسولین در بافت شکمبه گوسفند کرمانی

چکیده

هدف: یکی از مهم‌ترین گیاهان دارویی که دارای خواص ضد میکروبی، محافظ کبد و آنتی اکسیدانی بسیار قوی بوده و از زمان های بسیار قدیم مورد استفاده انسان قرار گرفته است رازیانه (Foeniculum vulgare mill) از خانواده Apiaceae است. نتایج مطالعات مختلف بر روی حیوانات اهلی ثابت کرده است که افزودن رازیانه در جیره غذایی حیوانات باعث افزایش هضم و بازده رشد می‌شود، کارایی لاشه و وزن و طول روده کوچک را بهتر می‌کند و تعداد کل باکتری‌ها را کاهش می‌دهد، و تبدیل خوراک و وزن بدن را بهبود می‌بخشد. لذا، هدف از این مطالعه بررسی تأثیر تغذیه رازیانه بر بیان ژن IGF1 در بافت شکمبه بره‌های کرمانی برای اولین بار بود.
مواد و روش‌ها: در این تحقیق از 30 راس بره نر از نژاد کرمانی با وزن 45/0 ± 5/27 کیلوگرم (هشت ماهه) استفاده شد. پس از پایان دوره مطالعه، حیوانات مورد بررسی برای نمونه برداری ذبح شدند و وزن شکمبه ثبت شد. RNA کل از بافت شکمبه استخراج شد و سپس cDNA سنتز شد. کیفیت RNA و cDNA با استفاده از الکتروفورز روی ژل آگارز اندازه گیری شد. از روش Pfaffl et al. برای تجزیه و تحلیل داده‌های حاصل از Real Time PCR استفاده شد.
نتایج: وزن شکمبه خالی در حیوانات تغذیه شده با رازیانه (780 گرم)، کمتر از حیواناتی بود که با رژیم غذایی بدون رازیانه (890 گرم) تغذیه شده بودند (P <0.05). RNA کل استخراج شده دارای کیفیت مطلوب، کامل، سالم و دارای دو باند با اندازه‌های S28 و S18 بود. الکتروفورز نتایج تکثیر ژن‌های هدف (IGF1) و مرجع (GAPDH) در بافت شکمبه روی ژل آگارز نشان داد که اندازه باند برای ژن هدف 265 جفت باز و برای ژن مرجع 76 جفت باز است. با افزایش سطح تغذیه با رازیانه میزان تکثیر ژن IGF1 در بافت شکمبه افزایش معنی داری (p<0.05) پیدا کرد. حداکثر بیان ژن IGF1 زمانی بود که حیوانات با جیره دارای سطح 2 درصد رازیانه تغذیه شدند.
نتیجه‌گیری: یافته‌های مطالعه حاضر نشان می‌دهد که رازیانه می‌تواند با تأثیر مفید بر بیان ژن IGF1 در شکمبه در جیره گوسفندان برای پیشرفت عملکردهای تولیدی (با تحریک رشد و نمو شکمبه) استفاده شود. اگرچه با توجه به یافته های پژوهش حاضر می‌توان تأیید کرد که رازیانه می‌تواند برای اهداف مختلفی در پرورش گوسفند استفاده شود، اما در تحقیقات آتی، به منظور دستیابی به یک نتیجه قطعی بهتر است شرایط فیزیولوژیکی و اپی‌ژنتیکی متنوع و پیچیده تری در نظر گرفته شود.

چکیده [English]

Fennel from the Apiaceae family is one of the most important medicinal plants that has strong antimicrobial, liver protective and antioxidant properties and has been used by humans since ancient times. The results of various studies on domestic animals have proven that adding the fennel in the diet of animals, increases digestion and growth efficiency, It improves carcass efficiency and small intestine weight and length , improves performance and health status, and improves feed conversion and body weight. Therefore, the aim of this study was to investigate the effect of fennel feeding on IGF1 gene expression in the rumen tissue of Kermani lambs for the first time.
Materials and methods
In this research, 30 Kermani male lambs weighing 27.5 ± 0.45 kg (eight months old) were used. After the end of the study period, the investigated animals were slaughtered for sampling and the rumen weight was recorded. Total RNA was extracted from the rumen tissue and then cDNA was synthesized. The quality of RNA and cDNA was measured using electrophoresis on agarose gel. The Pfaffl et al. method was used to analyze the Real Time PCR data.
Results
Empty rumen weight in animals fed with fennel (780 g) was lower than animals fed with diet without fennel (890 g) (P < 0.05). The extracted total RNA was of good quality, complete, and healthy, and had two bands with sizes of S28 and S18. Electrophoresis of the amplification results of target (IGF1) and reference (GAPDH) genes in rumen tissue on agarose gel showed that the band size for the target gene is 265 bp and for the reference gene is 76 bp. By increasing the level of feeding with fennel, there was a significant increase in IGF1 gene proliferation in rumen tissue (p<0.05). The maximum expression of IGF1 gene was when the animals were fed with a diet containing 2% fennel.
Conclusions
The findings of the present study show that fennel can be used with a beneficial effect on IGF1 gene expression in the rumen in the diet of sheep to improve production functions (by stimulating the growth and development of the rumen). Although according to the findings of the current research, it can be confirmed that fennel can be used for various purposes in sheep breeding, but in future research, in order to reach a definite result, it is better to consider diverse and more complex physiological and epigenetic conditions.
