بررسی اثرات ترکیبات مختلف هورمونی بر میزان باززایی مستقیم شاخساره در شرایط درون شیشه¬ای ریحان (Osmium basilicum L.)

چکیده

ریحان (Osmium basilicum) گیاهی علفی، متعلق به خانواده نعناعیان و غنی از اسانس­های روغنی است که برای اهداف مختلف دارویی، بهداشتی و خوراکی مورد استفاده قرار می­گیرد. مطالعۀ حاضر به منظور تعیین مناسبترین غلظت تنظیم کننده­های رشد اکسینی و سایتوکینینی و مشخص نمودن بهترین غلظت برای به دست آوردن بیشترین بازده در تولید گیاهچه­های درون شیشه­ای از ریزنمونه­های گره شاخساره گیاه ریحان در محیط کشت پایه MS انجام گرفت. آزمایش به صورت فاکتوریل در قالب طرح کاملاً تصادفی و با 3 تکرار اجرا گردید. در این آزمایش از سطوح مختلف تنظیم کنندۀ رشد BAP (0، 5/2، 5 و 5/7 میلی­گرم در لیتر) و IAA (0، 1/0 و 5/0 میلی­گرم در لیتر) استفاده گردید، و صفات درصد­باززایی، تعداد­باززایی در هر ریزنمونه، درصد نمونه­های شیشه­ای شده و میزان تولید ریشه اندازه­گیری و بررسی شد. بهترین باززایی از لحاظ درصد و تعداد ­باززایی در هر ریزنمونه، در محیطMS تکمیل شده با غلظت 5/2 میلی­گرم در لیتر BAP و بدون هورمون IAA به دست آمد. با افزایش غلظت هورمون BAP، میزان شیشه­ای شدن گیاهچه­ها افزایش یافت و بیشترین میزان ریشه­زایی در غلظت 5/0 میلی­گرم در لیتر IAA بدست آمد.

چکیده [English]

Basil (Ocimum basilicum ) is an annual, herbaceous plants belonging to the family of lamiaceae. Basil is used to as a herb, a spice as well as fresh vegetables. The present study aimed to investigate the effects of different concentrations of growth regulators such as auxin and cytokinin to identify the best hormonal concentration to obtain the highest yield of in vitro produced plantlets using nodal explants. Experiment was performed in factorial completely randomized design, using the basic MS. In this experiment the effects of different concentrations of BAP(0, 2/5, 5 and 7/5 mg/l) and indol acetic acid (0, 0/1 and 0/5 mg/l) was studied alone or together. Regenetaion percent, number of regeneration, vitrification and rooting evaluated. The results showed that the highest percentage and number of regeneration obtained in the medium included 2/5 mg/l BAP concentration and 0 mg/l IAA, respectively. Vitrification was increased with increasing BAP concentration, maximum rooting was observed in the medium containing 0/5 mg/l IAA.
